离体胰腺神经节的细胞内生物电记录及细胞电生理特性 被引量:1

摘要 应用细胞内记录研究猫离体胰腺神经节细胞的电生理特性。结果:细胞的静息电位为-58;5±8.7mV(下同);膜的输入阻抗(R_m)、时间常数(τ)和膜电容(C_m)分别为68.6±5.1MΩ、3.4±0.2ms和50.8±3.9pF;当细胞内注入去极化电流引起的电紧张电位达阈电位水平时,所有的细胞都能爆发锋电位,阈电位在静息电位基础上去极19.2±0.5mV,锋电位的幅度、超射值、时程分别为81.0±1.7mV、22.6±0.9mV和2.9±0.1ms。继锋电位后还出现一幅度为20.5±0.8mV的锋电位后超极化,刺激突触前神经时,在大部分细胞均能记录到快兴奋性突触后电位(f-EPSP)或顺向动作电位。f-EP-SP的幅值、时程及神经传导速度分别为8.9±0.7mV、25.8±1.9ms和0.48±0.04m/s。此外,在所有细胞都能观察到自发f-EPSP。f-EPSP由乙酰胆硷通过菸硷型受体引起。 The work was carried out to investi-gate electrophysiological properties of neurons of catintrapancreatic ganglia in vitro by means of intracellu- lar recordings .The mean value of resting membranepotential was-58.5±8.7mV with arange from- 45 to 72 mV, The mean values of mem- brane input resistance(R_m),time constant(τ)andcapacity(C_m) were 68.6±5.1MΩ,3.4±0.2ms and50.8±3.9pF (n= 28),respectively. When depolariz-ing electrotonic potentials induced by intracellular in-jection of depolarizing current pulses(0.05-0.5nA,5ms)reached the threshold potential level, all of theneurons could fire action potentials. The threshold.amplitude,overshoot and duration of spikes were 19.2±0.5mV, 81.0±1.7mV,22.6±0.9mV and 2.9±0.1ms(n=35).respectively.The spike was followedby a prolonged afterspike hyperpolarization with am-plitude of 20.5±0.8mV(n= 35). In most of neu-rons,fast excitatory postsynaptic potentials(f-EPSP)or orthodromic action potentials were recorded duringstimulation of nerve trunks attached to the intrapan-creatic ganglia. The mean values of amplitude,dura-tion of the f-EPSP and conduction velocity of thenerves were 8.9±0.7mV, 25.8±1.9ms and 0.48±0.04m/s(n=24),respectively. The ongoing synapticactivity was observed in all of cells. Moreover,f-EPSP.was induced by acetylcholine mediated through nico-tinic receptors.
