摘要
本文通过对耐高温酿酒酵母(S.cerevisiae)HU-TY-1产孢条件的研究,在菌龄、pH值、培养基组成,Zn^(2+)浓度及微量元素几个方面进行了实验。结果表明,HU-TY-1先在含2%葡萄糖,25μ/ml Zn^(2+)的预产孢培养基上培养,于稳定期前期转入含125~μg/ml Zn^(2+),1%KAc,0.25%酵母抽提物,0.1%葡萄糖,0.1%KH_2PO_4的产孢培养基上,可获得高的产孢率(45.92%)。另外,在产孢培养基中加入丁二酸等不被细胞代谢的物质作为缓冲剂,加入生物素,肌醇等维生素可大大提高菌株HU-TY-1产3-4孢(3-4孢即一个子囊中有3个或4个孢子)的能力。
Various affecting factors such as culture time,pH value,component of media,Zn concentration and trace elements on the sporulation of thermotolerant-yeast(Scerevisiae)HU-TY-1 were investigated. The result showed that cultivation in defined presporulation-medium containing 2% glucose and 25 g/mg Zn2+followed by sporulation in a medium containing 125g/mg/Zn2+,1% KAc,0.25% yeast extract,0.1 glucose and 0.1% KH2PO4 reached a maxmum sporulation rate of 45.92%. In addition,Bio-tine,Inositol and buffered sporulation medium consisted of an 0.2M solution;of Succinic acid(SUC) resulted in a further increase in yield of threerand four-spored asci.
