摘要
通过制备7264枚小鼠(Musmusculus)胚胎的染色体标本并鉴定其性别的试验,获得了理想的实验参数。采用理想的实验参数,制作和鉴定了242枚小鼠晚期桑椹胚、囊胚和扩展囊胚的染色体标本及其性别。性别鉴定成功率分别为80.4%、99.0%和94.5%。
An improved cytogenetic method for chromosomal preparation was used for sexing mouse (Mus musculus)embryos in the stage of late morula (LM)、blastocyst (B) and expanded blastocyst (EB). A total of 7264 mouse embryos were examined for sexing. Optimum parameters were found out: the concentration of colchicine, mitotic inhibitor, medium, the suitable development stage of mouse embryos for sexing, the duration of in vitro culture and the hypotonic treatment. 242 mouse embryos in LM、 B and EB were examined for sexing by using the optimum parameters, the sexing rates were 80. 4%、 99. 0% and 94. 5% respectively. Compared to the figures in published articles these results might be believed the best.
