摘要
本实验建立了小鼠脾细胞酸性α醋酸茶酯酶(ANAE)细胞化学染色方法。并采用ANAE染色方法检测了经羊红细胞(SRBC)免疫后的C57BL/6小鼠外周血及脾脏淋巴细胞染色结果,并对ANAE^+细胞进行了颗粒分型。结果表明,外周血中ANAE^+细胞数为69.0±5.4%,其中斑点型占54.5±5.0%,分散型占14.5±4.6%。小鼠脾脏ANAE^+细胞为43.1±12.1%,其中斑点型占35.7±10.6%,分散型占7.4±2.9%。同时观察了小剂量γ射线照射对小鼠外周血及脾脏淋巴细胞ANAE染色的影响,结果在24.4mGy至122.0mGy剂量范围内未见淋巴细胞亚群有明显变化。
A method of a-naphthyl acetate esterase (ANAE) staining for measuring lymphocyte subpopulations in spleen was established. It was shown that in normal mice immunized with sheep red blood cells (SRBC) ANAE+ cells in the spleen accounted for 43.1±12.1%, including 35.7±10.6% of the spot granular pattern and 7.4±2.9% scattered pattern, while ANAE+ cells in the peripheral blood accounted far 69.0±5,4%, including 54.5±5.0% of the spot granular pattern and 14.5±4.6% scattered pattern. Lymphocyte subpopulations were also examined in mice irradiated with γ-rays at a dose of 24.4 mGy to 122.0 mGy. No significant change was found between the irradiated and the controlled groups.
