摘要
以家兔骨骼肌组织提取细胞膜DHP受体的粗制品及提纯制品,经聚丙烯酰胺凝胶电泳。结果表明,骨骼肌细胞膜DHP受体蛋白亚单位的分子量为170、140、55和33k。采用3HPN200-ll0与DHP受体进行受体结合实验,其KD为2,5nmol/LBmax为58.9pmol/mg。人参单体皂甙Rb1表现出抑制DHP受体的作用,IC50为120ug/mi而人参单体皂甙Rg1则无明显作用。
The Dihydropyridine(DHP)receptors were purified from rabbit skeletal muscle transverse tubulemembranes using Chromatography on WGA-Sepharose,DEAE-Sephadex Chromatography and Sucrosegradient sedimentation. The Bmax and KD for 3H PN 200-110 binding to the T-tubule membranes are58.9 pmol/mg and 2.5 nmol/L respectively.The purified receptor proteins in SDS polyacrylamide gelelectrophoresis showed at least four polypeptides.These polypeptides have molecular weight of l70,140,55 ,and 33K. Ginsenoside Rb1 showed calcium antagonistic activity in the 3H PN200- 110 To theDHP recetpor binding assay.The IC50 of Ginssenoside Rb1 was l20 mg/ml but Ginsenoside Rg1 did notshow Calcium antagonistic activity in the same experiment
