摘要
将成年健康兔和大鼠的肾上腺髓质细胞移植至帕金森氏病大鼠模型的右侧侧脑室内。移植2个月后,兔肾上腺髓质移植组和鼠肾上腺髓质移植对照组分别有10只和5只动物的旋转行为得到明显改善。对这些旋转行为改善的动物均进行了脑的组织学检查。儿茶酚胺细胞的荧光组化法检查表明所有15只动物均见有含儿茶酚胺细胞的移植物,其中有些细胞仍保持正常细胞的形态特征,而另一些细胞则伸出粗短的突起。酪氨酸羟化酶抗体的免疫细胞化学染色显示有许多阳性细胞散布于移植区中,这些细胞多为圆形,无明显突起,部分为多形性细胞,有粗短突起出现。在尼氏染色的切片上没有发现淋巴细胞浸润的征象。以上结果表明移植的兔及大鼠肾上腺髓质细胞在帕金森氏病模型鼠脑内存活良好。本实验提示种系交叉的肾上腺髓质脑内移植是有可能的。
Implants of cells from adrenal medulla of adult rabbit and rat were placed into the right lateral ventricle of the 6-OHDA-lesioned rats. Rabbit and rat adrenal medulla were respectively grafted to 18 and 12 rats 10 and 5 of them had a significant improvement in rotational behaviour 2 months after grafting. All 15 recipients with rotational behaviour improved were perfused for histologic examinations. Catecholamine histofluorescence examinations revealed that all 15 animals had surviving grafts with catecholamine containing cells. These cells have undergone varying degrees of differentiation. Some cells retained a typical round appearence of normal adrenal chromaffin cells and a few cells grew into a more elongated form with short coarse processes. Immunocytochemical staining of tyrosine hydroxylase (TH) showed that there were many TH-positive cells within grafts. Some of them were rounded and had no obvious processes, and others were elongated or polygonal with short coarse processes, On the HE staining ions there were no obvious sign of lymphocytic in- filtration within or around graft areas. By esterase siaining, granular, diffuse esterase-positive and esterase-negative lymphocytes taken from all recipients' peripheral blood were counted at the 1st 3rd, 5th, 8th week after transplantation. The observation showed that transplantation of donor tissues from different species-of animals didn't cause obvious change in immunologic condition of graft recipients during two months. This study indicates that there is possibility of cross-species transplantation of adrenal medulla.
