摘要
本文对比了透析前后和在制备IL-1时加消炎痛对LPS诱导的腹腔细胞产生IL-1活性的影响。结果发现,在用LPS诱导巨噬细胞产生IL-1时,加入消炎痛可有效地去除IL-1上清中的环氧化酶代谢产物类抑制因子,且对IL-1检测系统无明显影响。此法简便实用,尤其适用于大量样品的检测。
In order to remove small inhibiting molecules such as prostaglandins in the fluid, IL-1 activity of LPS-stimulated peritoneal macrophage supernatants was commonly assayed after dialysis or high dilution. In the present study, we used indomethacin as cycloxygenase inhibitor, and found that IL-1 activity in indometh-acin + LPS-stimulated macrophage supernatant was higher than that without dialysis, but similar to that after dialysis. The results showed that indomethacin (10μM) could effectively inhibit prostaglandins production in the supernatant without any effect on IL-1 assay system itself.The method is simple and practical, and especially suitable for assaying large number of samples.
