六类十八种中药对血栓素A_2和前列环素合成的影响 被引量:16

摘要 本实验应用猪肺微粒体为供酶体,与花生四烯酸为底物进行反应,生成血栓素A_2(TXA_2)和前列环素(PGI_2)的方法,对6类18种中药进行实验研究。结果发现北沙参是一味对花生四烯酸代谢具有双向调节作用的药物;生大黄300mg组和灯盏花仅对TXA_2合成有明显抑制作用,对前列环素合成无明显影响;党参、黄芪、当归、人参皂甙、黄芩素等在一定剂量范围内主要抑制TXA_2合成,而对前列环素合成影响很小。这表明传统中药中存在着多种具有抑制TXA_2生成的中药,这些药物丰富了活血化瘀药的内容;而传统活血化瘀药在显著抑制TXA_2生成中仍占有重要地位。某些具有补益作用的中药具有不同程度的既抑制TXA_2合成,又促进或不抑制PGI_2合成的特点。这种扶正祛邪或祛邪不伤正的作用特点明显优于已知对照药阿司匹林和一些活血化瘀药。 The effect of 18 kinds of Chinese herbal medicine for the synthesis of TXA_2 and PGI_2 was studied. The porcine lung microsoma was used as donor of enzymes such as cyclo-oxygenase, thromboxane A2 synthetase and prostacyclin synthetase , etc. It was found that Glehnia littoralis could inhibit the synthesis of TXA_2 and increase the formation of PGI_2. Rheum palmatum (300mg group) and Erigeron breviscapus significantly inhibited the synthesis of TXA_2, but no apparent effection on the synthesis of PGI_2. When Codonopsis pilosulae, Astragalus membracaceus, Angelica sinensis, Ginsenosides and Baicalin, etc. markedly inhibited the formation of TXA_2 and mildly affected the formation of PGI_2. Some Chinese herbs for promoting blood circulation play an important role on the inhibition of TXA_2 synthesis. Some tonic herbs could either inhibit the synthesis of TXA_2 and increase the synthesis of PGI_2 or inhibit the formation of PGI_2. Therefore these tonic herbs had the characteristics of both strengthening the body resistance and eliminating pathogenic factors. In this aspect they are better than the control drug (Aspirin) and other herbs of promoting blood circulation.
