抗地高辛单克隆抗体制备及有关实验研究

摘要 以地高辛-人血清白蛋白偶联物为抗原,应用B细胞杂交瘤技术融合并筛选出7株分泌抗地高辛单克隆抗体的杂交瘤。该抗体可与地高辛人血清白蛋白特异性结合而不和其它具有类似化学结构的半抗原结合。用抗地高辛单克隆抗体治疗实验性地高辛中毒动物模型,可使地高辛中毒表现迅速消失。作者认为,抗地高辛单克隆抗体可作为地高辛的拮抗剂,对治疗地高辛中毒具有重要意义。 Digoxin was coupled with human serum albuminto produce antigen for immunization.7 ciones ofhybridomas secreting monoclonal antibody againstdigoxin-HSA were fused and screened through B cellhybridoma technique.This monoclonal antibody canapecifically combine with digoxin-HSA,but not withother semiantigens having analogously chemicalstruture.The ECG characters of digoxin intoxicationmay quickly disappear after the treatment of digoxinintoxication in exprnimental animal models withmonoclonal antibody to digoxin.The authors concludethat monoclonal antibody against digxion may be usedas an antagonist to digoxin which has important signif-icance in the treatment of digoxin intoxication.
