摘要
用抗弓形虫单克隆杂交瘤细胞株(3C_2)扩大培养,于腹腔注射BALB/C小鼠诱生腹水,经3次饱和硫酸铵盐析,透析,DEAE-Sephadex-A_(50)离子交换层析,除菌,制得纯化McAb(Ab_1),蛋白含量2mg/ml,用福氏佐剂乳化后免疫健康家兔4次,放血分离血清,再经盐析和层析处理,制得多克隆抗弓形虫独特型抗体(Ab_2),用佐剂制备疫苗,3次免疫绵羊(5～10mg/只)2只。共采集4次血清,制得抗抗弓形虫独特型抗体(Ab_3),用IHA法检测抗体效价为1:64～1024。证实抗独特型抗体(Ab_2)是特异的,是能模拟弓形虫抗原,具有类似弓形虫免疫原性,并可激发宿主免疫应答产生抗体,进而证实独特型和抗独特型的免疫网络学说在弓形虫病免疫中的应用价值。
Using against Toxoplasma McAb ( Ab1 ) immunity rabbit, poly against Id (Ab2) antibody is prepared. With which immunized health BALB/c mice, and challenged with (GJS) 104 of Toxoplasma virulent. The result showed that the unimmunity control group all died in 5 days, and immunity group can survival over II days. It's proved against Toxoplasma Id can induct immune response of mice and produce immunoprotection.
