摘要
本文应用逆转录聚合酶链反应(RT-PCR)方法检测乙型脑炎病毒基因。所设计引物在E基因组,引物1位于碱基序列的第1953~1972位,引物2位于第1788—1806位。反应产物为185bp,内含HaeⅢ限制性内切酶位点。产物在含溴化乙锭的2%琼脂糖中电泳。本法可特异性地检测乙型脑炎病毒核酸,并可检出少至5TCID_(50)的病毒RNA。
Reverse transcription--polymerase chain reaction (RT-PCR) was developed for the detection of Japanese encephalitis virus gene. Two primers bracketed a 186-nucleotide base in the E gene, corresponding to bases 1788-1972 in the Japanese encephalitis virus genomic RNA sequence. The amplified product including Hae Ⅲ site was shown in the stained 2% agrose gel by ethidium bromide. This method can specifically detect Japanese encephalitis viral RNA from at least 5TCID50 virus.
