摘要
本文在制备了华支睾吸虫全虫粗抗原(CsAg)、代谢抗原(EsAg)、表膜抗原(MAg)及其相应抗血清的基础上,应及石蜡和冰冻两种连续组织切片、IFAT、IGSS、PAP三种免疫化学方法法对华支睾吸虫的抗原定位进行了较系统的研究。结果表明:虫体定位器官主要是肠管、睾丸、储精囊和虫卵卵黄膜。两种切片抗原定位的主要区别是虫体皮层,石蜡切片未见皮层有抗原性,而冰冻切片虫体皮层有抗原性。在此基础上应用IE、SDS—PAGE研究了华支睾吸虫四种成虫抗原的免疫学特性,并研究了上述四种抗原的氨基酸组成,由此推测华支睾吸虫代谢抗原的生化特性可能以蛋白多糖为主。
Two kinds of serial section, cryostat sections and paraffin sections, three kinds of immuno-histochemical method including IFAT, IGSS and PAP were used to study systematically the localization of antigen of C. sinensis. The results showed that there were no obvious differences for localized organs among antisera of EsAg, MAg, CsAg and their IgG. The antigen formation organs mainly were worm's gut wall, testis, tegument seminal Vesticle and egg's vitelline membrane. The main difference between cryostat section and paraffin one was in tegument. Antigenicity was shown in the former but not in the latter. On the basis of antigen localization study, the biochemical and immunological characteristics of the worm's antigen were preliminarily analyzed by IE. SDS-PAGE and amino acid components analysis. According to the result of a series of experiments, the EsAg of C. sinensis may mainly be a proteoglycan.
