氟中毒雌鼠动情前期垂体催乳素细胞超微结构的变化

摘要 用喂饲含氟水的方法制造了雌性大鼠慢性氟中毒模型。以放免法测定了大鼠动情前期(Prestrus,P)期血清及垂体催乳素(Prolactin,PRL)水平,同时,在透射电镜下观察了P期大鼠垂体细胞超微结构的变化。结果是①氟中毒大鼠P期血清及垂体PRL水平均显著低于对照组。②电镜下氟中毒大鼠P期垂体PRL细胞呈现多种异常改变,主要表现为线粒体,粗面内质网等膜结构的损伤,同时胞质内成熟的分泌颗粒显著减少,仅见到少数未成熟的分泌颗粒。以上结果提示氟中毒时雌性大鼠P期PRL合成受阻。 Animal model of chronic fluorosis was developed in female rats drinking water containing sodium fluoride 100ppm for months. Serum and pituitary prolaetin were determined by RIA.The results were that both serum and pituitary prolactin level were decreased in proestrus rats with fluorosis. Ultrastructrue changes of lactotrophs in proestrus rats with fluorosis were as fellows: Most PRL cells showed the dence heterochromatins and swelling mitoehondria. RER dilated severely to form cisternel-like and the ribosome reduced.Mature secretory granules were greatly reduced,only some immature granules and secondary lysosome in cytoplasma.And deposition of fat droplet in cytoplasma can be seen. These results suggest that synthesis of prolactin was disturbanced in proestrus rats with fluorosis. And ultrastructure changes of lactotrophs may be involved in the possible mechanism of PRL alteration in proestrus rats with fluorosis.
