摘要
目的 制备甲胎蛋白 (AFP)特异性转移因子并鉴定其理化和生物学性质。方法 以基因重组AFP纯化后免疫猪 ,从猪脾脏离心超滤物中制备出AFP特异性转移因子 (TFAFP)。按《中国生物制品规程》规定检测理化性质 ,以Et RFc试验和白细胞移动抑制实验检测生物活性。结果 TFAFP各理化性质及生物活性均符合生物制品标准 ,并具有AFP抗原特异性。结论 TFAFP体外能转移对AFP的免疫性 ,可尝试用于肝癌治疗。
Objective To prepare and identify α-fetoprotein specific transfer factor (TF AFP). Methods By centrifugation and ultrafiltration, TF AFP was extracted from the porcine spleens, which had been immunized with recombined α-fetoprotein (AFP), bioactivities were detected with Et-RFc test and leukocyte migration inhibition test. Results Physicochemical properties and bioactivities of TF AFP were accorded with the standard of biological products, leukocyte migration inhibition test showed that TF AFP had AFP-antigen-specificity. Conclusion TF AFP can transfer the immunization to AFP in vitro, and can be tried to treat hepatocellular carcinoma.
