摘要
试验用6个苹果品种的子叶作外植体,比较了不同激素配比对诱导愈伤组织的作用以及对进一步分化的影响.试验表明用2,4-D 作为苹果子叶去分化的激素是有效的,但影响再分化.当2,4-D 与 KT 等配合使用可以提高愈伤组织的诱导率,但其愈伤组织进一步分化同样很难,苹果子叶对 NAA 极为敏感,但高浓度的 NAA 同样会抑制芽的分化.当低浓度的 NAA 与KT 配合能100%的诱导出愈伤组织,同时有不定芽的分化;而用6BA 与低浓度 NAA 配合,所诱导的愈伤组织具有更强的分化能力,能在短期内形成大量的不定芽,通过生根培养获得再生植株.
Cotyledons of six apple cultivars (Mulus pummila) were selected to be explants.The effect of different hormone combinations on the callus induction and differentiation were compared.The results indicated that 2,4-D accelerated dedifferentiation of cotyledon and re- strained formation of devntitious shoots.2,4-D—KT hormone combination increased callus induction rate,though the callus induced was difficult to differentiate.The response of cotyledon to NAA was sensitive,and the induction rate of low concentration NAA—KT combination was 100%,adventitious shoots could be formed in further differentiation. BA-NAA combination had the same effect on callus induction as KT-NAA.The callus in- duced had stronger regenerated ability,a lot of adventitious shoots could form in short period,and plantlet could be obtained through subculture in root-forming medium.
