摘要
TCP家族转录因子是植物所特有的转录因子家族,它主要在分生组织中起作用,与细胞分化和生长有很大联系。我们以哥伦比亚野生型拟南芥为模板得到基因At2g31070的全长ORF,生物信息学分析结果表明它具有TCP家族保守结构域,属于TCP家族的转录因子。以拟南芥成株不同组织的RNA为模板进行RT-PCR实验,结果发现较之其它组织,该基因在花中有比较高的表达量;通过酵母单杂交实验鉴定该基因具有转录激活活性。综合以上结果,我们认为At2g31070基因在拟南芥花形成发育的过程中发挥转录因子的作用,在一定程度上调控和影响了这个过程。
TCP family is a transcription factor family which particularly exists in plant genome. It mostly works in meristem and has great relationship with cell differentiation and cell growth. We cloned the full length ORF of gene At2g31070 from Columbia wild type Arabidopsis. According to the result of bioimformatics analysis, At2g31070 has the conserved TCP domain so belongs to TCP family. Using the cDNA from different tissues of adult Arabidopsis as templates, we got the result of RT-PCR which told us that gene expressed a higher level in flower than any other tissues. Afterward we proved that it had transcription activation function via yeast one-hybrid technique. All these results above showed us gene At2g31070 works as a transcription factor in the progress of Arabidopsis flower's growth.
