摘要
目的 观察奥曲肽对氧致小鼠视网膜新生血管形成的抑制作用,探讨其作用机制。 方法 20只新生小鼠进入密闭氧箱饲养5 d,在正常环境下继续饲养5 d。治疗组在结束给氧后给予奥曲肽50μg/kg腹腔注射每天2次,连续5 d,对照组给予等量生理盐水注射。出生第17 d时处死小鼠摘除眼球固定后切片,行HE染色计数突破内界膜的内皮细胞数,VEGF免疫组化检测视网膜VEGF表达。 结果 对照组平均每个切面突破视网膜内界膜的内皮细胞数为22个,治疗组平均为13个,两者相比差异有显著意义(P<0.01);治疗组VEGF染色较对照组减弱。 结论 奥曲肽能有效抑制氧致小鼠视网膜新生血管形成,有可能用于新生血管性疾病的治疗。
Objective To explore the effects of octreotide on retinal neovascularization and its related mechanism. Methods Twenty neonatal mice were exposed to 75% oxygen for 5 days and then to room an. Experimental mire were divided into two groups randomly. Each mouse in the treated group received twice mtraperitoneal injections of octreotide (50 μg/kg) per day for 5 days, and the control animals received the same dosage of BSS injection. All animals were sacrificed on post-natal day 17 and both eyes were enucleated. The active neovascular response was evaluated by counting endothelial cell nuclei of new vessels on the inner limiting membrane. The expression of retina vascular endothelial growth factor (VEGF) was detected with immunocytochemistry. Results About 22 neovascular nuclei per cross-section were seen in the treated group and less than 13 nuclei in the control group (P < 0. 01). Stain of retinal VEGF was weaker in the treated group than in the control group. Conclusion Intraperitoneal injection of octreotide can suppress retinal neovascularization effectively and decrease VEGF expression in the hyperoxia retina in part, suggesting a potential therapeutic effect of octreotide for retinal vascular disease.
