摘要
目的 观察缺血再灌注损伤对肾细胞游离钙 ([Ca2 + ]i)水平和细胞凋亡的影响。 方法 30只Wistar大鼠 ,分组建立急性缺血再灌注肾损伤模型 ,应用Fura 2 /AM荧光指示剂测定缺血再灌注大鼠肾细胞 [Ca2 + ]i水平 ,流式细胞术检测肾细胞凋亡率。 结果 缺血再灌注 1、2、2 4h肾细胞Ca2 + 超载水平分别为 15 6 .2、181.6和 2 6 0 .6nmol/L ,与对照组 (10 9.7nmol/L)相比差异有统计学意义(P <0 .0 1) ;1、2、2 4h肾细胞凋亡率分别为 9.5 8%、9.79%和 11.2 3% ,与对照组 (1.5 1% )相比差异有统计学意义 (P <0 .0 5 )。缺血再灌注细胞Ca2 + 超载与肾细胞凋亡率有正相关趋势 (r =0 .83,P >0 .0 5 )。 结论 缺血再灌注肾细胞呈现Ca2 + 超载 ,与肾细胞凋亡率呈正相关趋势 ,提示肾细胞钙超载可能是再灌注损伤肾功能障碍的重要原因。
Objective To study the effect of renal ischemic reperfusion injury on intracellular ionized calcium level and apoptosis. Methods The model of rat's acute renal ischemic reperfusion injury was established.A total of 30 Wistar rats were divided into 5 groups:control, 1 h ischemia and 1 h,2 h,24 h reperfusion groups (n=6 for each group).Renal intracellular [Ca 2+]i level was determined by Fura-2/AM fluorescence assay,and renal cell apoptosis rates were measured by flow cytometry. Results The renal intracellular [Ca 2+]i levels at 1h,2h and 24h timepoint of reperfusion were 156.2 nmol/L,181.6 nmol/L and 260.6 nmol/L,respectively,which were significantly higher (P<0.01) than control group (109.7 nmol/L).Apoptosis rates at 1 h,2 h and 24 h timepoint of reperfusion were 9.58%,9.79% and 11.23%,respectively,which were significantly higher (P<0.05) than control group(1.51%). Conclusions At different periods of rat’s acute renal ischemic reperfusion injury,renal intracellular [Ca 2+]i overloading is positively correlated with the rate of apoptosis,which suggests that overload of intracellular [Ca 2+]i may be an important cause of reperfusion injury.
