摘要
目的 :探讨酒精对锥体外系影响的不良机制。方法 :4 8只 8周龄雌性幼鼠随机分成对照组、低、中和高浓度组 ,分别用不同浓度的酒精 (0 %、12 .5 %、2 5 %和 5 0 % )灌喂 ,按 10 m l/kg,隔天 1次 ,90 d后 ,分别对各浓度组小鼠进行灌酒前、后转棒试验。转棒试验结束后快速取出小脑和尾壳核 ,并测量其丙二醛 (MDA)含量和超氧化物歧化酶 (SOD)活性。结果 :90 d后 ,高浓度组小鼠在继续灌酒前、后在棒上停留时间均减少 ,中浓度组小鼠灌酒后在棒上停留时间减少 ;高、中浓度组小脑和尾壳核的SOD活性降低 ,高浓度组小鼠小脑和尾壳核的 MDA含量升高 ,中浓度组小鼠小脑 MDA含量也升高。结论 :酒精致锥体外系功能障碍与小脑、尾壳核的 SOD活性降低和 MDA含量升高有关。
Objective:To explore the poisonous mechanism of etha nol exposure on the extrapyramidal system.Methods:48 mice(8 W ) were divided randomly into The control group, the low, middle and high dose g roups. Different doses ethanol (0%, 12.5%, 25% and 50%) were given (10 ml/kg) i n tragastrically by gavages once every other day for 90 days respectively. The rot arods were tested in 90 days before and after next ethanol administration. Mouse was sacrificed by decapitation after the rotarod test. The cerebellum and caudat e putamen were rapidly removed. SOD and MDA in the cerebellum and caudate putame n were assayed respectively.Result:The sustaining time of rota rod of The control group of the high dose group before and after exposure to eth anol were shorter in 90 days, and that of the middle dose group after exposure t o ethanol was also shorter. The activity of SOD in the cerebellum and caudate pu tamen of the high group decrease, while the content of MDA in the cerebellum and caudate putamen of the high group increase. the content of MDA in the cerebellu m of the middle group also decreased.Conclusion:There is a relation between the malfunctions of e xtrapyramidal system caused by ethanol and a decrease of SOD activity and an inc rease of MDA in cerebellum and caudate putamen.
