以 CEA 为靶抗原的基因免疫治疗是近年来肿瘤治疗学研究的突出进展,我们用 CEA 真核表达质粒(pCEA)免疫兔子,分别以重组人 CEA 痘苗病毒(rV-CEA)和生理盐水作阳性和阴性对照。与生理盐水相比,pCEA 组和 rV-CEA 组兔血清 CEA 含量均明显升高,并出现高滴度的作用时间持续更长久。
The immunogenicity of a DNA plasmid encoding the human full-length CEA cDNA, designated pCEA,was evaluated in rabbit.The rabbits were immunized with the pCEA,a recombinant human CEA-vaccinia virus (rV-CEA) or normal saline respectively.Serum CEA levels were elevated significantly in both pCEA-or rV-CEA-vaccinated rabbits,but the increasing serum CEA level lasted longer in pCEA treated rabbits than that in rV-CEA treated one.Anti-human CEA antibody was also detected in pCEA-or rV-CEA-vaccinated rabbit serum.The serum CEA and anti-human CEA antibody were not detected in normal saline treated rabbit serum.So the results show that,similar to the rV-CEA, pCEA also can highly express human CEA and induce a strong anti-human CEA response in rabbits, and the immune response induced by pCEA persisted longer than that by the rV-CEA.
