急性冠脉综合征患者MMP-3基因启动子5A/6A的多态性分析 被引量:4

The polymorphism of 5A/6A-MMP-3 gene promoter in acute coronary syndromes

摘要 目的 建立MMP 3基因启动子 5A/6A多态性的快速基因型分析方法 ,并且探讨此多态性对湖北武汉地区汉族人群急性冠脉综合征危险性的可能影响。方法 通过设计错配引物以及多聚酶链反应 限制性片段长度多态性 (PCR RFLP)分析方法 ,同时结合单链构象长度多态性 (SSCP)分析 ,检测了湖北地区10 3例汉族急性冠脉综合征患者及 10 0例健康成人的MMP 3基因 16 12 (5A/6A)的基因型。结果 急性冠脉综合征组与健康对照组MMP 3基因的 5A/6A +5A/5A基因型频率分别为 4 1 7%、 2 4 % ,5A等位基因频率分别为 2 1 8%、 13% ,两两比较均有显著性差异 (P <0 0 5 )。 5A/6A +5A/5A基因型的OR值为 2 2 6 9(95 %CI :1 2 4 1~ 4 14 9,P =0 0 0 7)。结论 MMP 3基因 5A/6A多态性与急性冠脉综合征危险性有关 。 Objective To establish a rapid method for analyzing the polymorphism of 5A/6A-MMP-3 gene promoter,and to investigate the possible impact of this polymorphism on the risk of acute coronary syndromes(ACS)in Han Chinese population.Methods 5A/6A polymorphism was detected in 103 Han patients with ACS and 100 healthy adults through polymerase chain reaction(PCR),restriction fragment length polymorphism(RFLP),and PCR-single strand conformation polymorphism(SSCP).Results The prevalence of 5A/6A+5A/5A genotype (41.7% vs 24%)and 5A allele(21.8% vs 13%)of MMP-3 gene were significantly more frequent in the ACS group than in control subjects(P<0.05).The odds ratio of the 5A/6A+5A/5A was 2.269(95%CI,1.241 to 4.149,P=0.007). Conclusion These findings indicate that the 5A/6A polymorphism of MMP-3 gene is associated with the risk of ACS,and might play an important role in the development of coronary atherosclerosis.
