一氟磷酸谷酰胺及αD_3对去势大鼠股骨、颌骨力学性质的影响 被引量:1

摘要 目的 :研究氟化物与αD3 联合用药对去势大鼠股骨、颌骨力学性质的影响。方法 :选 3月龄SD雌性大鼠分为假手术组、去势组、用药组。用药组于去势后 6周 ,用一氟磷酸谷酰胺 ( 0 .45mgF-+13 .5 6mgCa2 + /kg/d-1)与αD3 (αD3 0 .1ug/kg/d-1)灌胃 ,第 18周末结束实验。测血生化指标并作三点弯曲骨生物力学测量。结果 :①去势后大鼠体重增长迅速 ,较假手术组比较差异有显著性 (P <0 .0 5 )。②去势后雌激素下降明显 ,与假手术组比较差异有显著性 (P <0 .0 5 )。去势组碱性磷酸酶水平较假手术组升高 (P <0 .0 5 ) ,一氟磷酸谷酰胺与αD3 联合用药组碱性磷酸酶水平接近假手术组。③去势组股骨生物力学指标较假手术组没有明显变化 ,但氟化物与αD3 联合用药组力学指标较假手术组有较大提高。结论 :一氟磷酸谷酰胺组与去势组相比并不降低生物力学指标 。 Objective:The purpose of this study was t o investigate the effects o f monofluorphosphate and αD 3 on the mandibule and femur in ovariectomized rat s by three-point bending test.Method: Sprague-Dawley rats about 90 days of age we re randomly divided into the following groups and treated for 12 weeks: sham、o vx and treated groups. Animals of treated groups were received monofluorphosphat e for 6 days/week at a dose of 0.45 mg F- + 13.56 mg Ca 2+/kg/d -1 body weight, by gastric feeding at the beginning of seven weeks after ovariectomy.They were also treated withαD 3 at a dose of 0.1 μg/kg/d -1 body weight. All rats were sacrifise d by ether . The blood samples were collected for determination of biochemistry parameters. The left mandibule and fermur were preserved for three-point bendin g test. Result: ①Compared with sham group, ovariectomy was fol lowed by an inc rease in body weight.② The estradiol was obviously higher in the sham group t han in the OVX group. The alkaline phosphatase was significantly greater in OVX group compared with the sham group, however the alkaline phosphatase in MFP+αD 3 group was similer to that of sham group. ③There was no difference in biomec h anical index between OVX and sham, but the biomechanical index of MFP+αD 3 is be tter than that of the sham and OVX as well as MFP groups, respectively. Conclusi on: The combination of MFP with αD 3 increases the indices of biomec hanical strength over MFP treated alone. [
