溃疡性结肠炎组织中树突细胞的分布与TNF-α的表达 被引量:4

摘要 目的 :研究在溃疡性结肠炎组织中树突细胞的分布与TNF -α的表达 ,探讨溃疡性结肠炎的炎症免疫发病机制。方法 :收集 4 8例溃疡性结肠炎 ,5 0例大肠癌旁正常结肠粘膜组织标本 ,用免疫组化法检测肠粘膜内树突细胞的分布和TNF -α的表达。结果 :溃疡性结肠炎活动期树突细胞阳性率为 90 .6 0 % ,明显高于非活动期 (6 2 .5 % )和对照组 (30 .0 % ) ,组间比较P均 <0 .0 5。活动期TNF -α的阳性率为 87.5 % ,高于非活动期 (5 6 .3% )和对照组 (2 6 .0 % ) ,P均 <0 .0 5 ,树突细胞阳性率与TNF -α阳性表达呈正相关。结论 :树突细胞和TNF -α在溃疡性结肠炎肠粘膜组织中的阳性表达率 ,随疾病活动性增高 ,提示炎症免疫在溃疡性结肠炎发病机制中起重要作用。 Objective: To study the distribution of dendritic cells(DCs)and the expression of tumor necrosis factor-α(TNF-α) in mucosal tissue of ulcerative colitis (UC). Methods: Immunohistochemical method was used to detect the distribution of DCs and the expression of TNF-α in biopsy specimens of the colonic mucosa in 48 patients with UC and 50 normal tissues 10 cm away from colorectal tumor. Results: Positive rate of DCs was 90.60% in active UC, 62.5% in inactive group and 30.0% in controls, which was shown significant differences among the groups (P<0.05). Positive expression of TNF-α was 87.5% in active UC, 56.3% in inactive UC and 26.0% in controls. Distribution of DCs was positively related to expression of TNF-α in mucosal tissue in UC. Conclusion: DCs and TNF-α significantly increase in mucosa along with activity of UC and inflammatory immunity plays a key role in pathogenesis of UC.
