The significance of CRT_H2 expression on T cell subsets in patients with idiopa thic thrombocytopenic purpura

摘要 目的 研究CRTH2在特发性血小板减少性紫癜 (ITP)患儿外周血T淋巴细胞亚群的表达及其意义。方法 收集ITP患儿及健康儿童外周抗凝静脉血 ,分离纯化T细胞 ,以PE标记的抗CRTH2单抗和Cy5标记的抗CD4、CD8单抗作双色流式细胞术 ,分析CRTH2在ITP患儿CD4 + 、CD8+ T细胞表达的水平。结果 ITP患儿外周血T细胞与健康儿童相比 ,CD4 + 细胞百分率及CD4 + /CD8+ 比例明显下降 (P <0 .0 5 ) ,CD8+ 细胞百分率无明显变化 (P >0 .0 5 ) ;CD4 + CRTH2 -及CD4 + CRTH2 + 细胞百分率明显下降 (P <0 .0 5 ) ,CD8+ CRTH2 -及CD8+ CRTH2 + 细胞百分率无明显变化 (P >0 .0 5 ) ,CD4 +CRTH2 -/CD4 + CRTH2 + 及CD8+ CRTH2 -/CD8+ CRTH2 + 比例明显升高 (P <0 .0 5 )。结论 ITP患儿外周血存在T细胞亚群比例失衡 ,呈明显TH1类细胞优势 ,T细胞亚群比例失衡与该病的免疫发病机制有关。 Objective To determine CRT H2 expression on T c ell subsets in pediatric patients with idiopathic thrombocytopenic purpura (ITP ). Methods The ratios of CD4+/CD8+, CD4+CRT H2-/ CD4+CRT H2+ and CD8+CRT H2-/CD8+CRT H2+ in peripheral T cells wer e analyzed by immunofluorescence staining and bicolor flow cytometry (FCM). Results Compared with the ratios of CD4+CRT H2-/CD4+CR T H2+(47.21%± 5.36% ) and CD8+CRT H2-/CD8+CRT H2+ (20.47%±4.03% ) in healthy children, the ratios of CD4+CRT H2-/CD4+CRT H2+(61.18%±7.34% ) and CD8+CRT H2-/CD8+CRT H2+(25.06%±3.28%) in ITP children significa ntly increased. FCM analysis showed that the average percentages of CD4+, CD4 +CRT H2-, CD4+CRT H2+, CD8+, CD8+CRT H2- and CD8+CRT H2+ w ere 21.82%±6.5%, 21.48%±6.3%, 0.34%±0.16%, 33.14%±6.15%, 31.83%±5.48% and 1 .47%±0.97% in ITP children versus 39.03%± 5.6% , 38.24%±5.5%, 0.81%±0.19%, 33. 06%±5.63%, 31.52%±5.19% and 1.51%±0.68% in healthy children. The average perc entages of CD4+, CD4+CRT H2-, and CD4+CRT H2+ obviously decreased in children with ITP. At the same time, the average percentages of CD8+, CD8+C RT H2- and CD8+CRT H2+ did not changed. Conclusion The ratios of CD4+CRT H2-/CD4+CRT H2+ and CD8+CRT H2-/CD8+CRT H2+ in peripheral T cells increased in ITP children. It suggested that cellu lar immunity in ITP children is shifted to T H1 type immunity, which may play a n important role in the pathogenesis of ITP. [
