蛋白质组学研究进展及其在肿瘤中的应用

摘要 随着人类全基因组测序工作的完成 ,对基因功能即基因表达产物蛋白的研究已经拉开了序幕。蛋白质组学研究直接定位于蛋白质水平 ,大规模地分析组织细胞的蛋白质表达水平、翻译后修饰以及蛋白质间相互作用 ,是后基因组计划的重要组成部分。肿瘤的发生涉及一系列复杂的分子事件 ,从蛋白质整体水平上研究肿瘤 ,可以进一步揭示肿瘤发病的分子机制 ,为肿瘤的诊断、治疗、药物的筛选带来新的思路和途径 ,为肿瘤的研究提供新的平台。本文就目前蛋白质组学研究中的主要技术体系、策略及其在肿瘤研究中的一些进展作一综述。 Proteomics has become a hot spot in cancer research after the project HGP has been completed. Protemics aims at the large-scale analysis of protein levels in tissues and cells, including protein expression, post-translational modification and interaction of proteins. The tumor development involves a series of complicated molecular events, therefore, the study of tumors at protein level can further reveal molecular mechanisms of tumorigenesis and provides new ideas and approaches for the diagnosis, treatment, screening of drugs and a new platform for tumor research. This review focuses on the technical system, strategies and advances of proteomics researches.
