摘要
目的 :比较复方双黄连颗粒剂和黄芩单味药材提取物中黄芩苷在大鼠胆汁中代谢的差异。方法 :运用高效液相色谱法和液质联用技术检测黄芩苷的代谢产物种类和数量 ,并初步确定其结构。结果 :从给药后的大鼠胆汁中 ,发现了几个主要代谢产物并初步鉴定了其化学结构。黄芩颗粒剂和复方双黄连颗粒剂中黄芩苷经肝脏代谢后其主要的相同代谢物为 5 ,6 ,7 三羟基黄酮 6 O 葡萄糖醛酸苷 7 O 葡萄糖醛酸苷 ,5 ,6 ,7 三羟基黄酮 6 O 硫酸酯 7 O 葡萄糖醛酸苷和 5 ,7 二羟基 6 甲氧基黄酮 7 O 葡萄糖醛酸苷 ,胆汁中 ,黄芩单方代谢有原型黄芩苷和 5 ,6 ,7 三羟基 6 O 葡萄糖醛酸苷 ,而复方双黄连颗粒剂则无。结论 :黄芩颗粒剂与双黄连颗粒剂在黄芩苷的代谢产物存在明显差异 ,复方配伍促进了总黄芩素在胆汁中的排泄。
AIM: To compare metabolism of baicalin in Huangqin extract with that of compound Shuanghuanglian granules in rat bile. METHOD: Detecting the kinds and number of baicalin metabolites by HPLC-MS n and analyzing their structures. RESULT: The major metabolites were found in rat bile after oral administration and their chemical structures were primarily identified. The major same metabolites of baicalin of Huangqin extract and compound Shuanghuanglian granules in liver were baicalin-6-O-β-glucopyranuronoside-7-O-β-glucopyranuronoside,baicalin-6-O-sulfate-7-O-β-glucopyranuronoside and 6-O-methyl-baicalin-7- O-β-glucopyranuronoside. Baicalin and baicalin-6 -O-β-glucopyranuronoside were found in rat bile after Huangqin extracts were given orally but not in compound Shuanghuanglian granules. CONCLUSION: There were significant differences between metabolites in Huangqin granules and in Shuanghuanglian granules. The complex compatability promoted the bile excretion of the total baicalein.
