rhGH提高肾病综合征大鼠肾脏GHR表达

Effects of recombinant human growth hormone on renal GHR expression in nephrotic rats

摘要 目的 观察重组生长激素 (rhGH)对肾病综合征鼠 (NS鼠 )肾脏生长激素受体 (GHR)、胰岛素样生长因子 Ⅰ (IGF Ⅰ )及血清蛋白的作用。方法 制备阿霉素大鼠NS模型 ,设正常对照组 (6只 )、肾病组 (12只 )和rhGH治疗组 (12只 )。用酶联免疫吸附试验测尿微量白蛋白浓度 (ALB) ,S 丽春红测 2 4h尿蛋白定量 ,全自动生化仪测量血清总蛋白和白蛋白 ,以放射免疫法 (RIA)测血清IGF Ⅰ水平 ,以免疫组织化学法测肾脏GHR的表达。结果 ①NS组大鼠血清蛋白显著低于正常组 ;②NS组大鼠血清IGF Ⅰ浓度显著低于正常组 ;③NS组肾组织GHR显著低于正常组 ;④rhGH治疗 4周组肾脏GHR表达和血清白蛋白与肾病组同期相比显著上升。结论 rhGH通过提高肾脏GHR表达而提高血清蛋白的水平。 Objective To observe the role of rhGH therapy in serum albumin level , serum insulin-like growth factor (IGF-Ⅰ) and renal GHR expression in nephrotic rats. Methods Thirty male SD rats were randomly divided into control (n=6),doxorubincin-induced nephrotic (n=12) and rhGH-treated nephrotic groups(n=12). Urinary protein excreted in one day was measured with ponceau S. Albumin level in serum was measured with automatic biochemistry appliance. IGF-Ⅰ concentration in serum was measured with RIA . Renal GHR expression was measured by immunohistochem. Results Serum albumin and IGF-Ⅰand GHR expression levels were decreased in nephrotic rats compared with control rats . There were significant positive correlations between serum albumin level and renal GHR expression , and serum IGF-Ⅰ level and renal GHR expression .At the end of the fourth week , serum albumin level and renal GHR expression were significantly increased in rhGH-treated nephrotic group compared with nephrotic group. Conclusion rhGH can improve the serum albumin level by promoting the expression of renal GHR in nephrotic rats .
