摘要
目的 建立头孢他啶血药浓度的高效液相色谱 (HPLC)测定方法。方法 采用HPLC UV方法 ,内标为丙酮 ,色谱柱为 μbondaparC18(7 8mm× 30 0mm) ,流动相 :甲醇∶0 0 1mol/L磷酸二氢钾 (2 0∶80 ,V/V) ,检测波长 :2 5 4nm ,柱温 :室温 ,流速 :1 0ml/min。结果 头孢他啶的血药浓度在 10~ 12 0 μg/ml范围内与峰面积比有良好的线性关系 ,最低检测浓度为 0 1μg/ml,方法的平均回收率为 (98 0± 6 2 ) % ,日内变异系数≤ 6 8%。 结论 该方法简单 ,快速 ,准确 ,为头孢他啶的血药浓度监测提供了分析方法。
Objective To establish HPLC method to determine the serum concentration of ceftazidime.Methods A HPLC-UV assay was developed. The innerstandard was acetone. The determinate wavelength was 254 nm; The column was μ bondaparkC 18 ; The mobile phase was methnol-KH 2PO 4 (20∶80, V/V) and the column temperature was room temperature; The flow rate was 1.0 ml/min.Results The determination limit was 0.1 μg/ml. The ceftazidime was linear in the range from 10 μg/ml to 120 μg/ml. The recovery was no more than 6.8 %.Conclusion The HPLC method is simple, rapid, and accurate, and provides the reference methods for determination of serum concentration of ceftazidime.
