发根农杆菌转化胀果甘草的研究 被引量:5

摘要 利用发根农杆菌R1601对药用植物胀果甘草(GlycyrrhizainflatBat)进行转化,诱导其产生发根.在发根诱导过程中,分别用不同菌液浓度、不同浸染时间对胀果甘草子叶、胚轴进行转化处理,统计、比较各条件下的发根率,为发根进行液体悬浮培养以及随后的规模性培养提供数据基础.结果表明,用稀释2倍的菌液浸染子叶8min时,发根诱导率最高,为56.49%. The Glycyrrhiza inflat Bat was transferred, and was proceeded to induced the hairy roots growing using Agobacterium rhizgenses R1601 with Ri-plasmid. During the induction, Glycyrrhiza inflat Bat was transferred in different explants, different concentration of bacteria, different infecting period and different cultivating time. In the end, the hair roots induction efficiency of different explants in different systems was calculated to get some basic data about suspended cultivation for the hairy roots. Results showed that when taking the cotyledon as explant, the induction rate was highest if the cotyledon was infected for six or eight minutes by the bacteria which had been diluted four times.
