鼻NK/T细胞淋巴瘤生存素基因表达与细胞凋亡和增殖的关系 被引量:4

Study on Expression of Survivin Gene and Its Relations with Cell Apoptosis and Proliferation in Nasal NK/T Cell Lymphoma

摘要 目的 :探讨凋亡抑制基因生存素 (Survivin)在鼻NK/T细胞淋巴瘤中的表达 ,及其与细胞凋亡和增殖的关系。方法 :应用TdT介导的dUTP缺口末端标记 (TUNEL)技术和免疫组织化学链霉素抗生物素 过氧化酶连接法 (SP法 )检测 2 5例鼻NK/T细胞淋巴瘤组织和 10例反应性增生淋巴组织中的细胞凋亡、增殖细胞核抗原 (PCNA)和Sur vivin基因表达水平。结果 :2 5例鼻NK/T细胞淋巴瘤组织中Survivin表达阳性 17例 (6 8.0 0 % ) ,10例反应性增生淋巴组织中Survivin不表达 ,差异有显著性 (P <0 .0 1)。 2 5例鼻NK/T细胞淋巴瘤中平均凋亡指数 (AI)为 1.92 %± 0 .86 % ,平均增殖指数 (PI)为 4 1.4 8%± 5 .10 % ;Survivin表达阳性组AI(1.71%± 0 .6 1% )显著低于阴性组 (2 .2 9%± 0 .72 % ) ,PI(4 3.35 %± 4 .80 % )显著高于阴性组 (37.5 0 %± 2 .90 % ) ,差异均有显著性 (P <0 .0 5 ,P <0 .0 1)。结果 :鼻NK/T细胞淋巴瘤中Survivin基因表达上调 ,Survivin基因可能通过肿瘤细胞凋亡的抑制和细胞增殖参与NK/T细胞淋巴瘤的发生发展。 Objective: To investigate the expression of Survivin gene and its relations with cell apoptosis and proliferation in nasal NK/T cell lymphoma. Methods: TdT-mediated dUTP nick end labeling (TUNEL) and immunohistochemistry were used to study cell apoptosis, expression of proliferation cell nuclear antigen (PCNA) and Survivin in 25 nasal NK/T cell lymphoma and 10 reactive lymphoid tissues. Results: Survivin expression was positive in 17 NK/T cell lymphomas (68.00%) while no expression of Survivin was detected in the 10 reactive lymphoid tissues (P< 0.01 ). Apoptosis index (AI) and proliferaling index (PI) was 1.92%±0.86% and 41.48% ± 5.10 % respectively in nasal NK/T cell lymphoma. AI in nasal NK/T cell lymphoma cells with positive expression of Survivin was significantly lower than that with negative Survivin ( 1.71% ±0.61% vs 2.29%±0.72%) whereas PI was significantly higher (43.35%±4.80% vs 37.50%±2.90%). Both differences were significant (P<0.05 and P< 0.01 ). Conclusion: Survivin gene expression is up-regulated in nasal NK/T cell lymphoma. Survivin gene may play a role in the pathogenesis of NK/T cell lymphoma tumor by influencing cell apoptosis and proliferation.
