摘要
将大籽蒿(Artemisia sieversiana Ehrh.ex Willd.)地上部分乙酸乙酯提取物用硅胶柱层析分离和纯化,通过波谱方法鉴定化合物,用MTT法、单细胞电泳进行体外抗肿瘤活性研究.鉴定出4种化合物:洋艾素;Achillin;5,7,4’-三羟基-3’,5'-二甲氧基黄酮;3,5-二羟基-6,7,3’,4’-四甲氧基黄酮.MTT法结果表明洋艾素和Achillin对人肝癌细胞SMMC-7721的生长有较明显的抑制作用,且具有一定的时间和浓度依赖性。单细胞电泳显示对SMMC-7721细胞的DNA有损伤作用.所以洋艾素和Achillin对SMMC-7721细胞株的抑制活性与其DNA损伤有关.
Four compounds were isolated from the methanolic extract of the aerial parts of Artemisia sieversiana, and identified by means of chemical and modern spectral methods. Cell survival rate was determined by MTT, and DNA damage was estimated using single cell gel electrophoresis (comet assay). Absinthin and achillin showed inhibitory effects on SMMC-7721 cell lines in vitro and therefore, they might have certain anticancer activities by damaging DNA.
