摘要
为解决欧美杂种山杨成年树种快速繁殖的问题,并为研究原生质体分离、遗传转化和体细胞变异打下基础,我们建立了愈伤组织再生系统.其愈伤组织诱导最适培养基为WPM+6-BA 1.0 mg·L-1+NAA 0.1 mg.L-1、WPM+6-BA 1.0 mg·L-1,分化培养基为wPM+6-BA 03~1.0 mg.L-1,愈伤组织诱导率和分化率分别为90.67%和92%.
In order to develop a simple and feasible approach to achieve high frequency plant regeneration forPopulus tremula P. tremuloides, and to make a base for protoplast isolation, transformation, and somaclonalvariation study, the callus regeneration system was established. The most suitable medium for callus inductionof Populus tremula P. tremuloides was WPM supplemented with 1.0 mgL-1 6-BA and 0.1 mgL-1 NAA,WPMsupplemented with 1.0 mgL-1 6-BA, and for differentiation was WPM supplemented with 0.3~1.0 mgL-1 6-BA.The rate of callus induction and differentiation were 90.67% and 92% respectively.
