摘要
目的 确定一种对近交系大鼠遗传物质进行准确可靠、快速简便的遗传检测方法。方法 利用聚合酶链反应扩增技术对国内已知的 6个品系、8个近交系大鼠群体进行了DNA多态性的分析 ,并与生化标记分析进行比较。结果 (1)生化标记分析中除 2个群体SHR(哈 )和WKY(哈 )在Es 3位点出现可疑带外 ,其他位点均与国际标准相一致。 (2 )不同品系个体间具有多态性 ;同一群体不同个体之间除SHR(哈 )的SMST位点和WKY(哈 )的AGT位点出现一定的差异外 ,其他均没有差异 ;(3)不同地区、同一品系、不同个体之间也存在一定的差异。结论 微卫星DNA能有效地区分品系、品系与亚系 ,并能为大鼠遗传基因的研究提供一个准确可靠、快捷简便的检测方法。
Objective In order to set up the exact, convenient method in the heredity monitoring of inbred rats. Methods DNA polymorphisms of 8 colonies from 6 inbred rat strains were studied using PCR-analyzed microsatellites, and compared with biochemical marker analysis. Results (1)The result of biochemical markers analysis was coincident with imternational standard except Es-3 locus of SHR(Ha) and WKY(Ha). (2)There is a polymorphism among the various rat strains; and no polymorphism in the same rat strains except SMST locus of SHR(Ha) and AGT locus of WKY(Ha).(3)There is a little difference between the same inbred rat strains in different areas. Conclusion The method of PCR-analyzed microsatellites can be used for distingushing between inbred and outbred, different strains, strain and substrain, and it provides a speedy, convenient method for genetic research of the inbred rat.
