摘要
目的 建立成熟的小鼠体细胞核移植技术。方法 通过显微操作 ,以ICR鼠成纤维细胞核和C57鼠的去核卵母细胞进行胚胎的重构。重构胚经化学激活或 3种不同脉冲电压融合后再经体外或输卵管体内培养 ;3d后回收胚胎并进行胚胎的发育鉴定。结果 去核卵母细胞的成活率为 71 91%、化学激活率为 75 0 9%;刺激电压为 6 0V(80 μs、刺激 5次 )的融合率比其他几种方式高 ;经脉冲电压 70V(80 μs、刺激 2次 )处理后的胚胎发育潜力比其他几种方式处理后的胚胎发育潜力要好。结论 小鼠体细胞核移植技术基本获得成功。
Objective To develop nuclear transplantation of embryonic stem cells in mice.Methods Through microinjection, ICR mouse fibroblasts nucleus was transferred into the cytoplasm of C 57 mouse enucleated oocyte to reconstruct embryo by micromanipulation. The reconstructed embryos were treated with chemical activation or three different pulse electric currents, then cultured in oviduct or in vitro. The cultured embryos were collected three days later to identify embryonic development. Results 71.91% of transferred enucleated oocytes survived. Chemical activation rate was 75.09%.The embryonic fusional potential was the best by pulse current 60V(80μs, five times activation) than by pulse current 70V or 80V(80μs,any times activation),and the embryonic development potential was better by pulse current 70V(80μs, two times activation) than by pulse current 60V or 80V(80μs,any times activation).Conclusion The nuclear transfer method is with success basically.
