摘要
目的 观察大鼠胰腺冷缺血后细胞凋亡的变化。方法 大鼠胰腺肝素生理盐水原位灌注后切取并冷保存 0、3、6、12、2 4h ,分别用光镜、电镜、流式细胞仪及TUNEL法检测胰腺组织中细胞凋亡的变化。结果 冷保存6h胰腺组织中细胞凋亡数轻度增多 (P <0 .0 5 ) ,冷保存 12h及 2 4h时 ,细胞凋亡明显增多 (P <0 .0 1)。结论 胰腺冷缺血损伤可引起胰腺腺泡细胞凋亡。
Objective To investigate the impact of prolonged cold ischemia on pancreatic cell apoptosis in rats.Methods The pancreas was perfused with heparin saline. Tissue samples were taken and preserved at 4 ℃ to undergo cold ischemia for varied periods of time: 0, 3, 6, 12 and 24 hours.The pathologic changes of pancreatic tissue were studied under optic and electron microscopes. Apoptotic cells were identified by the terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) technique, and the apoptosis rate was assessed by flow cytometry.Results Apoptotic pancreatic cells increased slightly in the 6 h cold-ischemia groups(P<0.05), and became more prominent after 12 h and 24 h of cold ischemia (P<0.01).Conclusion Cold ischemia injury can induce pancreatic cell apoptosis.
