神经营养因子联用对老年痴呆鼠学习记忆能力和基底前脑小白蛋白的影响

摘要 目的:探讨Neurturin和神经生长因子(nervegrowthfactor,NGF)联合应用对老年痴呆鼠学习记忆能力的影响以及与基底前脑小白蛋白(parvalbumin,PV)神经元的关系。方法:切断老年SD大鼠左侧穹窿海马伞(fimbria-fornix,FF),侧脑室注射Neurturin和NGF,用Y迷宫测试大鼠的学习记忆能力。用免疫组化结合图像分析技术观察Neurturin,NGF对大鼠基底前脑PV阳性神经元数目的影响。结果:损伤组大鼠空间学习记忆能力明显下降(P<0.01);NGF组学习记忆能力均有改善(P<0.05),但与正常组比较差异有显著性意义(P<0.05);联合组的学习记忆能力与正常组差异显著无显著性意义(P>0.05)。大鼠的学习记忆能力与基底前脑PV阳性细胞数呈正相关。结论:Neurturin和NGF的联合应用较单独使用NGF更好改善老年FF损伤鼠学习记忆能力。 AIM:To study the relationship of neurturin and nerve growth factor(NGF) unite application on abilities of learning and memory in rats with Alzhemizer's disease(AD). METHODS:AD model rats with left fimbria fornix(F F)transection received continous infusion of NGF and neuriturin into lateral ventricle.Learning and memory abilities were measured by Y maze test. The number of basal forebrain Parvalbumin positive neurons was analyzed by using immunohistochemical method combined with technique of micromeasure and image analysis. RESULTS:Remarkable decrease in spatial learning and memory abilities was found in the injured group(P< 0.01).The rats in the NGF group had significant improvement in learning and memory abilities(P< 0.05), while significant difference was found between the NGF and normal control groups(P< 0.05),but not between the combination group and normal control group(P >0.05).The abilities of learning and memory were positively related with the number of Parcalbumin positive cells. CONCLUSION:Combination treatment of nruruturin and NGF is more effective in improving the ability of spatial learning and memory in model rats with FF transection as compared with NGF alone.
