抗细粒棘球绦虫成虫单克隆抗体的制备及鉴定 被引量:3

摘要 目的 建立能分泌与细粒棘球绦虫 (简称包虫 )成虫特异结合的抗包虫成虫单克隆抗体的杂交瘤细胞株。方法 用包虫成虫抗原免疫BALB/c小鼠后 ,获取免疫的脾细胞 ,与小鼠骨髓瘤细胞SP2 / 0融合。结果 ELISA方法筛选出 1株能持续稳定分泌抗包虫成虫单克隆抗体的杂交瘤细胞株 ,1F5E3C9E9D5杂交瘤细胞株分泌的单克隆抗体可以与包虫成虫、棘球蚴结合 ,与囊液抗原呈边缘性阳性反应 ,而与泡球蚴EM2抗原呈阴性反应。冻存 1个月后复苏 ,仍能稳定分泌。经免疫组织化学检测发现 ,该单克隆抗体与棘球蚴的生发层 ,棘球蚴原头节虫体呈阳性反应。结论  1F5E3C9E9D5是一株稳定的杂交瘤细胞株 ,所分泌的抗细粒棘球绦虫成虫单克隆抗体在粪抗原的检测方面有应用前景。 To obtain a monoclonal antibody against adult worm antigens of Echinococcus granolosus,the mouse spleen cells immunized with the soluble antigens of the adult worms were fused with murine SP2/10 myeloma cells.After screening,a hybridoma cell strain steadily secreting monoclonal antibody against adult worm antigens of E.granolosus was obtained and named as 1F5E3C9E9D5. This antibody could react with adult worm antigens and the cyst fluid at edge, but not with the alvecococcus antigen EM2. The hybridoma cells could secret specific antibody steadily after one month of freezing and thawing. By immunohistochemical assay, it was found that the antibody reacted with the antigens in the germinal layer and protoscolex. It concludes that 1F5E3C9E9D5 is a stable hybridoma cell strain to secret monoclonal antibody against the adult worm antigen of E.granolosus, and this monoclonal antibody may be a good candidate for the detection of adult worm antigens in the stool of dogs.
