以钾离子透入引起的大鼠甩尾反应为痛指标,观察了电和化学刺激视上核(SON)对大鼠痛(?)(PT)和电针(EA)镇痛的影响。电刺激SON后,PT明显高于假刺激组(P<0.05~0.001),电刺激SON后电针足三里,镇痛效应明显提高,并有明显的量效关系。电刺激SON的近旁部位(0.5~1 mm)对PT及电针镇痛无明显影响。SON内注射L-谷氨酸(L-Glu)后痛(?)和电针镇痛效应都明显高于对照组,也有明显的量效关系。侧脑室注射纳络酮(Naloxone,NX)对SON内注射L-Glu的镇痛效应无明显影响,但可部分减弱电针镇痛效应。上述结果提示,下丘脑视上核在痛觉调制和电针镇痛过程中发挥重要作用,这种作用可能主要是由非阿片肽类物质介导的。
The effects of electro-and chemo-stimulating (ES, CS) supraoptic nucleus (SON) on analgesia and electroacupuncture (EA) analgesia were observed using potassium inotophoresis induced tail-flick in rats. The results showed that: different electrical current (12.5 ~ 50 μA) stimulating SON elevated the PT (P<0.05 or 0.01 vs sham stimulating group), while ES in combination with electroacupuncture 'Zusanli' increased the PT significantly (P<0.05-0.001, compared with sham stimulating-EA group). The effects of ES SON correlate with the current in the range of 12.5 ~ 50 μA. The stimulation of area nearby SON did not affect PT and the effect of EA analgesia. The injection of L-glutamic acid (L-Glu) into SON also produced similar effect on PT and analgesia by ES SON. Naloxone failed to block the effect of stimulating SON on PT with L-Glu, but could block enhancing EA analgesia with L-Glu stimulating SON partially. These results suggested that the hypothalamic supraoptic nucleus play an important role in regulating pain and EA analgesia. This function might be mediated mainly by non-opioid substance.
