摘要
合成了甲基对硫磷的半抗原,并与载体蛋白偶联合成了人工抗原,然后通过免疫的方法制备了其单克隆抗体。在筛选甲基对硫磷单克隆抗体的基础上,建立酶联免疫检测方法,并研制出检测苹果、玉米、蔬菜中甲基对硫磷残留的试剂盒。该试剂盒的灵敏度为0.5μg/L,对苹果、玉米、蔬菜样品的最低检测限分别可达到10、20、50μg/kg;加标回收率为70.3%~102.4%,变异系数为7.4%~13.0%;对实际样品的检测结果与气相色谱法基本一致。该方法快速、灵敏、可靠,为水果、蔬菜、谷物中甲基对硫磷的残留检测提供了便捷、准确的分析手段。
A hapten artificial antigen was prepared,and monoclonal antibodies against parathion-methyl were produced.On the basis of parathion-methyl monoclonal antibody,an enzyme-linked immunosorbent assay(ELISA) kit for parathion-methyl residues in apple,corn and vegetable was developed.The sensitivity of the ELISA kit was 0.5 μg/L.The detection limits for apple,corn,vegetable were 10,20,50 μg/kg,respectively.Sample recoveries were 70.3%~102.4%,and the coefficients of variation were 7.4%~13.0%.Results of the ELISA kit were in accordance with those by gas chromatography(GC).This method is rapid,sensitive and reliable,which provides a convenient and accurate way for detection of parathion-methyl residues in fruit,cereal and vegetable.
