蝴蝶兰不同耐热性品种幼苗对高温胁迫的生理反应 被引量:30

Response of Physiology of Phalaenopsis Cultivars with Different Heat Tolerance to High Temperature Stress at Seedling Stage

摘要 为了探明高温胁迫下蝴蝶兰不同品种耐热性差异的原因,以耐热性存在差异的2个蝴蝶兰品种幼苗为材料,分别在25℃、35℃、40℃各处理2天,测定了抗氧化酶活性、膜脂过氧化、质膜透性和渗透调节物质的变化。结果表明,随着温度的上升,2个品种的相对电导率和丙二醛(MDA)含量均不断增加,而耐热性强的品种的相对电导率和丙二醛含量增幅明显小于耐热性弱的品种;随着温度的上升,2个品种的超氧化物歧化酶(SOD)、过氧化物酶(POD)活性、可溶性蛋白含量均为先升而后降,但耐热性强的品种的SOD、POD活性、可溶性蛋白含量增幅明显大于耐热性弱的品种,而降幅明显小于耐热性弱的品种;随着温度的上升,2个品种的脯氨酸和可溶性糖含量均持续增加,而耐热性强的品种的脯氨酸和可溶性糖含量增幅明显大于耐热性弱的品种。由此得出,耐热性强的品种具有较强的细胞渗透调节能力、蛋白质合成能力和蛋白质稳定性以及消除活性氧(ROS)伤害的能力,是其耐热性较强的生理基础之一。SOD活性、POD活性、MDA含量、相对电导率、可溶性蛋白含量、可溶性糖含量、脯氨酸含量,可以作为蝴蝶兰不同品种间耐热性鉴定的指标。 In order to elucidate the causes of different heat tolerance of different Phalaenopsis cultivars under high temperature stress,changes of antioxidative enzyme activities,membrane lipid peroxidation,cell membrane permeability and osmotic adjustment substances of two Phalaenopsis cultivars seedlings with different heat tolerance which were treated at 25℃,35℃,40℃ for two days respectively were determined.The results showed that the electrolytic leakage and contents of malondialdehyde (MDA) in two Phalaenopsis cultivars with different heat tolerance were increased continuously with high temperature going up.However,the growing rate of the electrolytic leakage and contents of MDA in cultivar of heat resistance was smaller than that in cultivar of heat sensitivity.The superoxide dismutase (SOD),peroxidase (POD) activities and solube protein content show a transient rise and then reduce with high temperature rising,but the growing rate of the SOD,POD activities and solube protein content in cultivar of heat resistance was larger than that in cultivar of heat sensitivity,the range of decrease the SOD,POD activities and solube protein content in cultivar of heat resistance was smaller than that in cultivar of heat sensitivity.The contents of free proline and soluble sugar were increased continuously with high temperature going up.However,the growing rate of the contents of free proline and soluble sugar in cultivar of heat resistance was larger than that in cultivar of heat sensitivity.The above results indicated that there were better capability in maintaining cell osmotic adjustment,synthesising and stabling protein and eliminating reactive oxygen species (ROS) in cultivar of heat resistance,this is one of all physiological base which cultivar of heat resistance had high heat tolerance.The SOD,POD activities,MDA content,electrolytic leakage,solube protein,soluble sugar and free proline contents were selected as physiochemical indexes of evaluating on heat tolerance of different Phalaenopsis cultivars.
