siRNA对人阴茎海绵体平滑肌细胞5型磷酸二酯酶表达的抑制作用 被引量:3

摘要 目的观察小干扰RNA(siRNA)对人阴茎海绵体平滑肌细胞5型磷酸二酯酶(PDE5)表达的抑制作用,为阴茎勃起功能障碍(ED)的基因治疗提供实验依据。方法使用美国Ambion公司提供的设计软件设计人PDE5基因的siRNA序列,并合成3对PDE5siRNA和1对阴性对照siR-NA,转染人阴茎海绵体平滑肌细胞。RT-PCR法半定量检测siRNA对PDE5mRNA表达的抑制作用;Westernblot法检测海绵体平滑肌细胞PDE5蛋白表达水平。结果siRNA1、siRNA2和siRNA3转染后人阴茎海绵体平滑肌细胞PDE5mRNA表达分别下降58.2%、14.9%和11.9%;PDE5蛋白表达量下降约70.5%、19.8%和17.3%;阴性对照siRNA组未引起PDE5mRNA和蛋白表达的变化。结论体外化学合成的PDE5siRNA能特异有效地下调PDE5基因表达,不同序列特异性的siRNA下调PDE5基因表达的能力不同,为ED的基因治疗提供了新思路。 Objective To investigate the inhibitive effect of siRNA(small interfering RNA,siRNA) on the phosphodiesterase type 5(PDE5) in smooth muscle cells of human corpus cavernosum,and provide experimental groundwork for the gene therapy of erectile dysfunction (ED).
