摘要
目的探究维生素D(vitamin D,VitD)对系统性红斑狼疮(systemic lupus erythematosus,SLE)鼠外周血相关指标的影响。方法将6只C57/B6小鼠(普通小鼠)随机分为A组和B组,12只MRL/lpr小鼠(SLE模型鼠)随机分为C、D、E、F组,每组3只。其中A、F组给予100μl丙二醇溶液灌胃;B组小鼠以VitD 10 ng/(kg·d)灌胃;C组小鼠以VitD 10 ng/(kg·d)灌胃;D组小鼠以VitD 30 ng/(kg·d)灌胃;E组小鼠以VitD 10 ng/(kg·d)+帕立骨化醇0.8μg/kg灌胃,3次/周。灌胃6周后运用酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测各组小鼠炎症指标:肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)、新蝶呤(neopterin,Npt)、降钙素原(procalcitonin,PCT)、超敏C反应蛋白(hypersensitive C-reactive protein,hs-CRP)、单核细胞趋化蛋白1(monocyte chemotactic protein 1,MCP-1);免疫指标:抗核抗体(antinuclear antibody,ANA)、抗双链DNA抗体(anti-double-stranded DNA antibodies,dsDNA)、抗Sm抗体、免疫球蛋白G(immunoglobulin G,IgG)、补体C3;骨代谢指标:25-羟维生素D[25-hydroxyvitamin D,25(OH)D]、甲状旁腺素(parathyroid hormone,PTH)、成纤维细胞生长因子23(fibroblast growth factor 23,FGF23)。结果与A组比较,F组各项炎症指标显著上升;与F组比较,B、C、D、E组炎症指标显著降低,组间比较差异均有统计学意义(P均<0.01)。与F组比较,C组ANA、dsDNA、抗Sm、补体C3水平降低,对IgG无明显影响;D组ANA、dsDNA、抗Sm水平进一步降低,增加IgG、补体C3表达;E组ANA、dsDNA、抗Sm水平降至最低,组间比较差异均具有统计学意义(P<0.01)。与F组比较,C、D、E组25(OH)D、PTH水平降低,FGF23水平增加,组间比较差异均具有统计学意义(P<0.01)。结论VitD能改善SLE小鼠的炎症状况、免疫及骨代谢,与帕立骨化醇联用效果更佳。
Objective To investigate the effect of vitamin D(VitD)on peripheral blood indexes of systemic lupus erythematosus(SLE)mice.Methods Six C57/B6 mice were randomly divided into group A and B,and 12 MRL/LPR mice were randomly divided into group C,D,E,F,respectively,each group were 3 mice.Both group A and F were given propylene glycol solution(100μl)by gavage;the mice in group B were given VitD[10 ng/(kg·d)]by gavage;the mice in group C were given VitD[10 ng/(kg·d)]by gavage.The mice in group D were given VitD[30 ng/(kg·d)]by gavage.The mice in group E were given VitD[10 ng/(kg·d)]+Paricalcitol(0.8μg/kg)by gavage,3 times per week.Inflammatory indicators such as tumor necrosis factor alpha(TNF-α),neopterin(Npt),procalcitonin(PCT),hypersensitive C-reactive protein(hs-CRP),monocyte chemotactic protein 1(MCP-1);immune indicators such as antinuclear antibody(ANA),anti-double-stranded DNA antibodies(dsDNA),anti-Sm,immunoglobulin G(IgG),C3 and the bone metabolism indicators such as 25-hydroxyvitamin D[25(OH)D],parathyroid hormone(PTH),fibroblast growth factor 23(FGF23)of mice in each group were tested by enzyme linked immunosorbent assay(ELISA)after six weeks.Results Compared with group A,the inflammatory indexes of group F were significantly increased;compared with group F,the inflammatory indexes of groups B,C,D,E were significantly decreased(all P<0.01).Compared with group F,the levels of ANA,dsDNA,anti-Sm and C3 of group C were significantly reduced(P<0.01),while no significant difference was discovered in IgG;the levels of ANA,dsDNA and anti-Sm of group D were further reduced(P<0.01),and the expression of IgG and C3 were significantly increased(P<0.01).Compared with group F,the levels of 25(OH)D and PTH of group C,D,E were significantly reduced(P<0.01),and the level of FGF23 were significantly increased(P<0.01).Conclusion VitD can improve the inflammation,immunity and bone metabolism of SLE mice.Combining with paricalcitol has a better effects.
