人IP3R蛋白的分子结构和理化性质分析 被引量:1

摘要 目的为进一步研究肌醇1,4,5三磷酸受体(IP3R)3种亚型蛋白调控机制及生物学功能提供理论基础。方法利用NCBI、Protscale、TMHMM等数据库和生物信息学分析工具,对人IP3R 3种亚型蛋白的结构、性质、功能域及蛋白质空间结构进行分析。结果人IP3R 3种亚型具有跨膜结构,均为亲水性蛋白,含有大量α-螺旋,定位于内质网,具有多个磷酸化位点和跨膜结构,无二硫键,有离子通道活性和钙释放通道活性,参与离子转运并与多种蛋白存在相互作用关系等。结论人IP3R蛋白在参与和调节离子转运、细胞死亡、能量代谢等许多生理过程中起着重要的作用。 Objective To contribute theoretically to further research on regulatory mechanisms and biological functions of inositol 1,4,5-triphosphate receptors(IP3Rs).Methods Modern bioinformatics online databases and analytic tools,including NCBI,Protscale,TMHMM and SignalP,were used to analyze the possible structure and potential functions of human IP3R protein.Results The three subtypes of human IP3R were hydrophilic proteins that contained a large number ofα-helices and were located in the endoplasmic reticulum.Moreover,these proteins had multiple phosphorylation sites and transmembrane structures,but had no disulfide bonds.They had ion channel activity and calcium release channel activity,participated in ion transport and interacted with various proteins.Conclusion Human IP3Rs play an important role in participating in ion transport,cell death,energy metabolism and many other physiological processes.
