平目汤抑制Graves眼病小鼠眼眶脂肪积聚的研究 被引量:3

Study of Pingmu Decoction( 平目汤) on Accumulation ofOrbital Adipose in Mice with Graves' Orbitopathy

摘要 目的建立Graves眼病动物模型,观察平目汤对造模雌性BALB/c小鼠眼眶脂肪积聚的影响,并探讨其作用的分子机制和发挥作用可能的靶点。方法通过使用重组h-TSHR质粒基因免疫结合电穿孔的方法,建立Graves眼病动物模型。使用平目汤不同剂量及地塞米松灌胃4周,观察小鼠眼球后脂肪组织占眼球重量百分比、血清TRAb、甲状腺与眼球组织病理。结果(1)平目汤不同剂量及地塞米松干预造模小鼠后,小鼠眼球后脂肪组织占眼球重量百分比由低到高依次为:空白组<平目汤中剂量(PMT-M)组<平目汤低剂量(PMT-L)组<地塞米松组<平目汤高剂量(PMT-H)组<模型组。平目汤不同剂量组与地塞米松组均较模型组下降(P<0.05),PMT-L组与PMT-M组较地塞米松组和PMT-H组下降更加显著(P<0.05)。(2)小鼠血清TRAb水平由低到高依次为:空白组<PMT-M组<PMT-L组<地塞米松组<PMT-H组<模型组。与空白组相比,除PMT-M组外其他组别有统计学意义(P<0.05);与模型组相比,平目汤不同剂量与地塞米松干预后TRAb明显下降(P<0.05);与地塞米松组相比,PMT-L组与PMT-M组下降显著(P<0.05),其中PMT-M组更加明显。(3)眼球组织HE染色:与模型组相比,平目汤各组眼球后脂肪组织堆积减少,地塞米松组无明显改变。(4)甲状腺组织HE染色:模型组可见甲状腺淋巴细胞浸润,间质增生明显。平目汤不同剂量干预后淋巴细胞浸润减少,地塞米松干预后甲状腺少量淋巴细胞浸润。结论平目汤通过有效抑制Graves眼病小鼠眼球后脂肪组织的积聚而发挥疗效。 Objective An animal model of Graves’Orbitopathy was established to observe the effect of Pingmu Decoction(平目汤)on the accumulation of orbital adipose in the female BALB/c mice and explore the molecular mechanism and the possible target of the action.Methods The animal model of Graves’Orbitopathy was established by immunorecombination of human TSHR recombinant plasmid gene combined with electroporation.Different doses of Pingmu Decoction and dexamethasone were administered separately by gavage for 4 weeks to observe the percentage of the weight of posterior eye tissue adipose in the total weight of the eyeball,the serum TRAb,histopathology of the thyroid and eyeballs.Results(1)After different dosages of Pingmu Decoction and dexamethasone intervention in model mice,the percentage of the weight of posterior eyeball adipose tissue in the total weight of the eyeball was(from low to high):blank grou P<middle dose group of Pingmu Decoction(PMT-M)<low dose group of Pingmu Decoction(PMT-L)<dexamethasone group<high dose group of Pingmu Decoction(PMT-H)<model group.Those of the Pingmu Decoction groups and dexamethasone group were decreased compared with those of the model group(P<0.05).There was a significant decrease in PMT-L group and PMT-M group compared with dexamethasone group and PMT-H group(P<0.05).(2)The levels of serum TRAb in mice were(from low to high):blank group<PMT-M group<PMT-L group<dexamethasone group<PMT-H group<model group.Compared with those of the blank group,those of the other groups except PMT-M group were statistically significant(P<0.05).Compared with those of the model group,after the intervention of the different doses of Pingmu Decoction and dexamethasone,TRAb was significantly decreased(P<0.05).Compared with the dexamethasone group,PMT-L group and PMT-M group had the most obvious decrease(P<0.05).The latter was relatively more obvious.(3)HE staining of eyeball tissues:Compared with that of the model group,posterior eye tissue adipose accumulation was decreased in Pingmu Decoction groups,while that of dexamethasone group had no significant changes.(4)HE staining of thyroid tissue:Thyroid lymphocyte infiltration was observed in the model group and the interstitial hyperplasia was obvious.Different doses of Pingmu Decoction reduced the infiltration of thyroid lymphocytes after intervention,while after dexamethasone intervention,a small amount of lymphocyte infiltration of the thyroid gland can be observed.Conclusion Pingmu Decoction can effectively inhibit the accumulation of adipose tissue in the eyeballs of BALB/c mice.
