针刺“人中”“内关”对脑出血大鼠血肿周围脑组织NGB、Cyt-C表达的影响 被引量:8

Effect of Acupuncture on Expressions of NGB and Cyt-C in Brain Tissues around Hematoma of Cerebral Hemorrhage Rats

摘要 目的观察大鼠脑出血(ICH)急性期血肿周围脑组织脑红蛋白(NGB)、细胞色素C(Cyt-C)的表达随时间动态变化及针刺干预对其影响,探讨针刺"人中""内关"对大鼠ICH急性期的神经保护及细胞凋亡的影响。方法健康雄性SD大鼠96只随机分为对照组、模型组、穴位组、非穴组,每组24只。采用自体非肝素抗凝动脉血二次注入大鼠尾状核制作动物模型,符合纳入标准的大鼠再按出血后6、24、48、72 h分为4个时间点亚组,每组6只大鼠。穴位组采取针刺"人中""内关"干预;非穴组针刺非穴点干预;对照组及模型组捆绑束缚,干预后对应时间点取材。采用NSS评分法对术后大鼠神经功能缺损程度进行评分;运用免疫组化法定性测定血肿周围脑组织NGB、Cyt-C蛋白表达情况。结果与模型组相比,穴位组大鼠神经功能缺损体征明显降低,在72 h差异有统计学意义(P<0.05)。与模型组相比,穴位组各时间点NGB表达均增高(P<0.05)、Cyt-C表达均下降(P<0.05);与穴位组相比,非穴组各时间点NGB表达低于穴位组(P<0.05)、Cyt-C表达高于穴位组(P<0.05)。结论针刺"人中""内关"能改善ICH大鼠神经功能缺损体征,还可能通过提高出血后血肿周围脑组织NGB表达、降低Cyt-C表达,从而保护脑组织,抑制神经元凋亡,拮抗出血后继发的神经元损伤。 Objective To observe the dynamic changes of expressions of NGB and Cyt-C in brain tissue around hematoma of cerebral hemorrhage rats over time and the effects of acupuncture on themandexplore the effects of acupuncture on nerve protection and apoptosis in acute stage of cerebral hemorrhage rats.Methods A total of 96 healthy male SD rats were randomly divided into four groups:Control,Model,Acupoints and Non-acupoints,24 rats in each group.The model of ICH rats was established by autologous blood injection.According to 6,24,48 and 72 h after hemorrhage,they were divided into 4 subgroups,6 in each group.The acupoints group used acupuncture atNeiguan(PC6)and Renzhong(DU26).In the non-acupoints group,non-acupoints were stimulated.The control and model group used binding intervention and the samples were collected at the corresponding time points after intervention.NSS was used to evaluate the degree of nerve function defect in postoperative rats.The expressions of NGB and Cyt-C were measured by immunohistochemistry.Results Compared with those of the model group,the signs of neurological deficit in the acupoint group were significantly lower,and the difference was statistically significant at 72 h(P<0.05).Compared with those of the model group,the expression of NGB was increased(P<0.05)and the expression of Cyt-C was decreased(P<0.05)in the acupoint group at each time point.Compared with that of the acupoint group,the expression of NGB in the non-acupoint group was lower than that in the acupoint group at each time point(P<0.05),and the expression of Cyt-C in the non-acupoint group was higher than that in the acupoint group(P<0.05).Conclusion AcupunctureatRenzhong(DU26)and Neiguan(PC6)can improve the signs of neurological deficit in ICH rats.It may also protect brain tissue,inhibit neuronal apoptosis and antagonize secondary neuronal injury after hemorrhage by increasing the expression of NGB and decreasing the expression of Cyt-C in brain tissue around hematoma after hemorrhage.
