氢溴酸樟柳碱对大鼠空间学习记忆能力的影响 被引量:2

摘要 目的探讨氢溴酸樟柳碱对空间学习记忆能力的影响。方法将40只SD大鼠随机分为4组:阴性对照组、氢溴酸樟柳碱低、中、高剂量组。通过尾静脉注射给药,阴性对照组给予0.9%氯化钠注射液,氢溴酸樟柳碱低、中、高剂量组分别给予0.3mg、3mg、15mg·kg-1氢溴酸樟柳碱注射液,连续给药14d。采用Morris水迷宫试验评价氢溴酸樟柳碱对大鼠空间学习记忆能力的影响,并定期监测动物体质量变化。结果各组大鼠体质量在整个给药期呈现相似的增长趋势,组间比较差异无统计学意义。与阴性对照组比较,氢溴酸樟柳碱中、高剂量组能显著增加第1天上午训练时的上台潜伏期和上台前路程(P<0.05,P<0.01);随着训练次数的增加,各剂量组大鼠在训练后期,潜伏期及路程均低于阴性对照组,其中高剂量组在第2天下午的潜伏期,中剂量组和高剂量组分别在第3天上午、第2天下午的上台前路程,与阴性对照组比较显著降低(P<0.05)。站台撤除后,氢溴酸樟柳碱各剂量组大鼠的站台穿越次数和站台周围路程与阴性对照组相比差异无统计学意义。结论氢溴酸樟柳碱中、高剂量能干扰大鼠短期空间学习记忆能力以及促进长期空间学习记忆能力,对大鼠的空间学习记忆具有双相作用。 Objective To investigate the influence of anisodine hydrobromide on spatial learning and memory abilities of rats.Methods 40 SD rats were randomly divided into four groups:Negative control group,low,medium,high level dose group of anisodine hydrobromide.Through tail vein injection,0.9%saline was giving to negative control group,0.3 mg,3 mg,15 mg.kg-1 anisodine hydrobromide was giving to low,medium,high level dose group respectively.The whole administration duration lasted 14 days.Morris water maze was used to evaluate the Influence of anisodine hydrobromide on spatial learning and memory abilities,And the weight of rats was weighed regularly.Results During the whole duration of administration,the weight of rats was increased steady in each group,none significant differences were observed.Compared with control group,pre-stage incubation period and pre-stage distance of first-day morning can be significantly increased in medium,high level dose groups(P<0.05,P<0.01).With the increasing of training times,pre-stage incubation period and pre-stage distance were all lower than control group in each administration group,among which,pre-stage incubation period in the second day afternoon of high level dose group was obviously shorter than control group(P<0.05),pre-stage distance in the third day morning and the second day after of medium and high level respectively were both shorter than control group(P<0.05).After removing the stage,number of stage-crossing and round of the stage in each administration group had no significant differences comparing to the control group.Conclusion Medium and high-level dose group of anisodine hydrobromide can hinder the shortterm ability of spatial learning and memory,on the contrary,long-term ability of spatial learning and memory can be enhanced.Hence,the influence of anisodine hydrobromide on spatial learning and memory abilities was biphasic.
