Upon all catalysts tested, 4-TBC is themain product and 3-tert-butyl catechol (3-TBC) and 3,5-di-tert-butyl catechol are the sideproducts. The synthetic PMHs showed higher conversion of catechol and better selectivity to 4-TBCcompared to other solid acid catalysts tested. Over the PMHs derived from H-exchangedmontmorillonite through template extraction processes, the suitable reaction temperature is ca 410K, the ratio of catechol to tert-butyl alcohol is 1:2. Increasing the amount of catalyst (lowerweight hourly space velocity) can improve the conversion of catechol and influence the selectivityslightly. The reasonable reaction time is ca 8 h. The type and strength of acidity ofH-montmorillonite and PMH were determined by pyridine adsorption FT-IR and ammoniatemperature-programmed desorption techniques. The medium and strong acid sites are conducive toproducing 4-TBC and the weak acid sites to facilitating the 3-TBC formation. The differences betweenthe PMHs from calcination and those from extraction are attributed to proton migration and aciditychange in the gallery surface.展开更多

Improving the nutrition of laying hens is a crucialmeasure to reverse this declination. Thisstudy investigatedthe effect of seleniumyeast (SY)on egg production, ileal gene expressionandmicrobiota, aswell as elucidating their associations in aged laying hens. A total of 375 Jinghong laying hens at 76weeks oldwere randomly assigned into 5 dietary treatments, which included a selenium-deficient basal diet based oncorn-soybean meal, and dietary supplementation of SY at 0.15, 0.30 and 0.45 mg/kg, and sodium selenite at0.45mg/kg.The results showed that SYamelioratedthe depressionin aged layingperformance inthe 0.30mg/kg group (P < 0.01). Selenium yeast significantly increased ileum selenium concentration (P < 0.05), and SYgroups had higher seleniumdeposition efficiency than the sodiumselenite group. Functional enrichment andShort Time-series Expression Miner (STEM) analysis indicated that SY activated metabolic progress (e.g.,glycerolipidmetabolism, glycerophospholipidmetabolism, andfattyacidmetabolism),immune responseandoxidative stress response. Four hub genes including thioredoxin reductase 1 (TXNRD1), dihydrolipoamidedehydrogenase (DLD), integrin linked kinase (ILK) and leucine zipper tumor suppressor 2 (LZTS2) wereinvolved in intestinal metabolismwhich was closely associated with selenium deposition/status. Moreover,the relative abundance of Veillonella, Turicibacter and Lactobacillus was significantly increased, but the relativeabundance of Stenotrophomonas was significantly decreased by SY supplementation. Multi-omics dataintegration and Canonical correspondence analysis (CCA) showed that both the ileumselenium content andthe laying rate were highly correlated with pathways and bacteria enriched in metabolism and immuneresponse. Meanwhile, the “switched on” gene prostate stem cell antigen (PSCA) had a positive relationshipwith Veillonella and a negative relationship with the opportunistic pathogens Stenotrophomonas. Overall, ourstudy offered insight for the further exploration of the role of SY on boosting egg production and balancingileum intestinal flora in aged laying hens.展开更多
In the current study,the Zygomycetes fungus Cunninghamella elegans NRRL Y-1392 was evaluated for its ability to grow in extracts derived from dried and ground agricultural residues,such as mushroom stalks and roots fr...In the current study,the Zygomycetes fungus Cunninghamella elegans NRRL Y-1392 was evaluated for its ability to grow in extracts derived from dried and ground agricultural residues,such as mushroom stalks and roots from hydroponically cultivated lettuces and produce poly-unsaturated fatty acids(PUFA)andγ-linolenic acid(GLA)rich lipids.Initially,the compositions of stalks and lettuce roots were analysed,and the fungus was batch-flask cultivated on six different commercial semi-defined substrates containing different sugars detected in stalks and roots to evaluate its catabolic ability.C.elegans was capable to assimilate all sugars,but at a lower rate in the case of arabinose.Subsequently,C.elegans was cultivated on tailor-made semi-defined commercial substrates,resembling hydrolysates containing carbohydrates found in mushroom stalks,under both nitrogen-excess and nitrogen-limited conditions,and resembling that of hydrolysates of roots,under nitrogen-excess conditions.Based on the results,under nitrogen-excess conditions,in the case of media resembling stalks hydrolysates,higher production values for biomass,PUFAs,and GLA were observed(20.3 g/L,1906 mg/L,668 mg/L),accompanied by high productivity values due to short cultivation periods,while under nitrogen limitation,high lipid accumulation(lipid in dry cell weight=48%,w/w)was presented,and lipids rich in oleic acid were produced.Finally,the fungus was cultivated on a medium derived from hot water-extraction applied to mush-room stalks,enriched with organic nitrogen sources.The fungus was successfully grown on the sugar-rich waterextract derived from mushroom stalks,resulting in dry biomass of 14.5 g/L,lipids of 1.8 g/L,with 15%(w/w)of GLA in cellular lipids.展开更多
The natural curcumin-mediated photodynamic inactivation(PDI)was developed,and its inactivation potency against Fusarium graminearum in vitro and in vivo was systematically investigated by fluorescence probe assay,tryp...The natural curcumin-mediated photodynamic inactivation(PDI)was developed,and its inactivation potency against Fusarium graminearum in vitro and in vivo was systematically investigated by fluorescence probe assay,trypan blue staining,scanning electron microscope(SEM),confocal laser scanning microscopy(CLSM),etc.Results showed that under the irradiation of blue LED,the photosensitizer of curcumin was excited to generate massive reactive oxygen species(ROS)in the cells of F.graminearum,and the PDI completely inactivated their mycelia and spores under the treatment of 150μM curcumin and 10.8 J/cm^(2)irradiation.Further analysis found that the PDI ruptured the cellular microstructures,damaged the cell membrane by increasing its permeability and oxidizing the lipids,degraded the intracellular DNA and proteins inside the spores of F.graminearum.Meanwhile,the PDI also potently killed>99.99%spores of F.graminearum on maize under the treatment of 200μM curcumin and 10.8 J/cm^(2)irradiation.Moreover,the PDI suppressed the production of zearalenone(ZEN),and residual ZEN could not be detected after the storage of maize for 10 days.Therefore,this study systematically explored the inactivation efficiency of curcumin-mediated PDI against both the mycelia and spores of F.graminearum,which provides a valid and promising method to control the fungal hazards in grains.展开更多

The ai...Toxicity is the totality of adverse effects, which can be functional and morphological lesions in a living organism, caused by a substance introduced in relatively high single doses or in small, repeated doses. The aim of this study was to assess the OECD-recommended acute oral toxicity and anti-inflammatory activity of ethanolic extract of Ficus umbellata leaves. Animals were given a single oral dose of 1000, 3000 and 5000 mg/Kg body weight (BW) of the extract. For the anti-inflammatory activity test, rats were given the ethanolic extract of F. umbellata leaves at doses of 100, 300 and 500 mg/Kg or aspirin® at a concentration of 100 mg/Kg PC orally, one hour before injection of 0.05 ml of 1% formalin under the plantar fascia of the rat’s right hind paw. Paw volume measurements were taken one, two and three hours after formalin injection, using an electronic caliper. After 14 days of observation, no deaths were observed in treated rats. The LD50 of ethanolic extract of Ficus umbellata leaf powder is greater than 5000 mg/Kg body weight. This extract has no significant effects on hematological parameters and on the main markers of nephrotoxicity and hepatotoxicity for a single dose of less than 5000 mg/Kg PC. It reduces formalin-induced edema. Evaluation of the percentage inhibition showed that the extract had greater anti-inflammatory activity at 3 hours after the start of the experiment. However, better inhibition of inflammatory oedema of the paw of rats treated with 500 mg/Kg was observed at 5 hours after the start of the experiment, with a percentage inhibition of 69.23 ± 1.02, compared with the reference group treated with aspirin® 100 mg/Kg, which showed an inhibition of 63.50 ± 0.98. These results show that F. umbellata leaves possess anti-inflammatory activity, which would justify their use in traditional African medicine to prevent or treat inflammation.展开更多
