Objective:To explore the protective effect and possible mechanism of action of Zhuyu Annao pill in mice with intracerebral hemorrhage(ICH).Methods:Sixty mice were divided into the control group,hemorrhage group,drug-t...Objective:To explore the protective effect and possible mechanism of action of Zhuyu Annao pill in mice with intracerebral hemorrhage(ICH).Methods:Sixty mice were divided into the control group,hemorrhage group,drug-treated group(after hemorrhage),TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group(after hemorrhage) with 12 in each group.Model of autologous ICH was established in all groups.After drilling and 12 h of fasting,models in the control group hemorrhage group and TLR4-knockout hemorrhage group were all drenched with 10 mL/kg distilled water by intragastric administration.Models in the drug-treated group and TLR4-knockout hemorrhage + drugtreated group were drenched with 6.25 g/kg of Zhuyu Annao pill.All groups were treated for 7 d.Longa scoring method was used to measure the neurological defect scores and determine the brain water contents of all groups;ELISA was employed to detect the inflammatory factor interleukin(IL)-6,tumor necrosis factor- α(TNF- α) and IL-1β in brain tissues;and Western blot was applied to test the expression quantities of apoptotic protein Bax and anti-apoptotic protein Bcl-2 in brain tissues.Results:At day 3 and7,compared with the hemorrhage group,the neurological defect scores of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group decreased significantly(P<0.05) Compared with the hemorrhage group,the brain water contents of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group reduced significantly(P<0.05) Compared with the hemorrhage group,the inflammatory factor IL-6,TNF-α and IL-1β of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group decreased significantly(P<0.05).Compared with the hemorrhage group,the expression of apoptotic protein Bax of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage+ drug-treated group decreased significantly and the expression of anti-apoptotic protein Bcl-2 increased significantly(P<0.05).Conclusions:Zhuyu Annao pill can alleviateencephaledema for mice with ICH and reduce inflammatory responsesandnerve cell apoptosis.TLR4 can mediate inflammatory injury induced by ICH.Thus,Zhuyu Annao pill can play a protective role for brains by decreasing the expression of TLR4.展开更多

X-ray diffraction(XRD) results indicate that incorporating MWCNTs into...Poly(vinylidene fluoride)(PVDF)/multi-walled carbon nanotube(MWCNT) nanocomposites were prepared by means of ultrasonic dispersion method. X-ray diffraction(XRD) results indicate that incorporating MWCNTs into PVDF caused the formation of β phase. A thermal annealing at 130 ℃ confirmed that the β phase was stable in the nanocomposites. Differential scanning calorimetry(DSC) results indicate that the melting temperature slightly increased while the heat of fusion markedly decreased with increasing MWCNT content. The tensile strength and modulus of PVDF were improved by loading the MWCNTs. The scanning electron microscopy(SEM) observations showed that MWCNTs were uniformly dispersed in the PVDF matrix and an interfacial adhesion between MWCNT and PVDF was achieved, which was responsible for the enhancement in the tensile strength and modulus of PVDE.展开更多

The contents of malondialdehyde (MDA), reduce...The effect of H2O2 pretreatment on Cd tolerance and translocation of rice seedlings were studied using two rice cultivars (N07-6 and N07-63) differing in Cd tolerance. The contents of malondialdehyde (MDA), reduced glutathione (GSH), non-protein thiols (NPT), phytochelatins (PCs) and the activity of glutathione S-transferase (GST) were compared between the two cultivars exposed to various treatments. The results showed that 50 μmol/L Cd exposure significantly inhibited rice growth, enhanced the production of GSH, NPT, PCs and MDA, and increased the activity of GST, and there were significant differences between the two cultivars. More Cd was transported into the shoot of N07-6. The H2O2 pretreatment alleviated Cd toxicity by further increasing GSH, NPT and PCs contents, as well as the GST activity in roots. The increase degrees of these parameters in N07-63 were higher than those in N07-6, suggesting that the tolerance of N07-63 was enhanced more significantly than N07-6. Hydrogen peroxide reduced Cd translocation to rice shoot but affected the Cd content in root differently. From the above results, it may be speculated that there were remarkable differences in the Cd detoxification and response to the H2O2 pretreatment between the two cultivars.展开更多
Background:Alzheimer’sdisease(AD)is a prevalent neurodegenerative disorder causing progressive dementia.Research suggests that microRNAs(miRNAs)could serve as biomarkers and therapeutic targets for AD.Reduced levels ...Background:Alzheimer’sdisease(AD)is a prevalent neurodegenerative disorder causing progressive dementia.Research suggests that microRNAs(miRNAs)could serve as biomarkers and therapeutic targets for AD.Reduced levels of miR-137 have been observed in the brains of AD patients,but its specific role and down stream mechanisms remain unclear.This study sought to examine the therapeutic potential of miR-137-5p agomir in alleviating cognitive dysfunction induced in AD models and explore its potential mechanisms.Methods:This study utilized bioinformatic analysis and a dual-l uciferase reporter assay to investigate the relationship between miR-137-5p and ubiquitin-specific peptidase 30(USP30).In vitro experiments were conducted using SH-SY5Y cells to assess the impact of miR-137-5p on Aβ1-42 neurotoxicity.In vivo experiments on AD mice evaluated the effects of miR-137-5p on cognition,Aβ1-42 deposition,Tau hyperphosphorylation,and neuronal apoptosis,as well as its influence on USP30 levels.Results:It was discovered that miR-137-5p mimics efficiently counteract Aβ1-42 neurotoxicity in SH-SY5Y cells,a protective effect that is negated by USP30 overexpression.In vivo experiments demonstrated that miR-137-5p enhances the cognition and mobility of AD mice,significantly reducing Aβ1-42 deposition,Tau hyperphosphorylation,and neuronal apoptosis within the hippocampus and cortex regions.Mechanistically,miR-137-5p significantly suppresses USP30 levels in mice,though USP30 overexpression partially buffers against miR-137-5p-i nduced AD symptom improvement.Conclusion:Our study proposes that miR-137-5p,by instigating the downregulation of USP30,has the potential to act as a novel and promising therapeutic target for AD.展开更多

The results showed that as the concentration of nitrobenzene increased, the growth of N. hantzschia was inhibited, and the algal culture liquids gradually changed from dark yellow to light yellow after 5 d of treatment;the soluble sugar content increased after 2 d;and the soluble protein content of the 100 mg/L nitrobenzene treatment group was 89.1% of the control group on day 1, which was the lowest value, and then showed a gradual upward trend. The low-mass concentration of nitrobenzene promoted the chlorophyll a content of algal cells, the medium and high mass concentrations had an inhibitory effect, and the chlorophyll a content of the 50 mg/L treatment gradually recovered after 3 d. The electrical conductivity of algal cells gradually increased with the increase of the mass concentration of nitrobenzene. The electrical conductivity gradually recovered after 3 d of the low mass concentration treatment, while the high mass concentration harmed the algae cells, causing N. hantzschia to gradually die.展开更多

In the absence of Sl endonuclease, the fluorescence signal of the ROX-ssDNA was efficiently quenched when the ROX-ssDNA was adsorbed on the surface of AgNRs via strong electrostatic interaction. However, upon addition of different concentration of Sl endonuclease, the fluorescence was gradually restored owing to the reduction of FRET efficiency caused by S1 endonuclease specific cleavage ROX-ssDNA into short fragments, which reduced the electrostatic interaction between AgNRs and short oligonucleotide fragment. This assay strategy exhibited a high sensitivity and excellent specificity for S1 endonuclease with a detection limit of 0.004 U/mL and a dynamic concentration range from 0.01 U/mL to 5.0 U/mL. In addition, the capabilities for screening of S1 endonuclease inhibitors and S1 endonuclease detection from complex biological matrixes were also verified.展开更多

Methods: Rat thyroid of different age (10, 45, 65, 100, 150 weeks) was isolated and thyrocytes cultured. Total RNA was extracted in different rat age group when thyroid cells had been cultured for two weeks. mRNA IGF-1 expression was measured with reverse-transcription polymerase chain reaction( RT-PCR) in each group and compared. Results: Quantity of total RNA in thyroid cells decreased with ageing when the rat thyroid cells had been cultured for 2 weeks. There is significant difference among groups (P<0.05). Expression of IGF-1 mRNA could be detected in thyroid cells of different age cultured in vitro. Quantity of IGF-1 mRNA expression by RT-PCR analysis increased from 10 to 45 weeks old, and then decreased with ageing. Conclusion: Rat thyroid cells from different age cultured in vitro can express IGF-1 mRNA. Quantity of total RNA in thyroid cells cultured in vitro decreased with aging. IGF-1 mRNA expression was correlated to age (r=0.401, P<0.05).展开更多

Objective:To investigate the effects and mechanisms of tetrahydroberberine(THB)on vascular endothelial cell injury induced by oxygen glucose deprivation(OGD)and LPS-induced cell injury model.Methods:MTT assay was used...Objective:To investigate the effects and mechanisms of tetrahydroberberine(THB)on vascular endothelial cell injury induced by oxygen glucose deprivation(OGD)and LPS-induced cell injury model.Methods:MTT assay was used to detect the effects of THB final concentration on the survival rate of EA.hy926 cells,which were 0μg/mL,2.5μg/mL,5μg/mL,10μg/mL,20μg/mL and 40μg/mL,respectively.Group I of the experiment:normal control group,OGD group,OGD+THB-10μg/mL group,OGD+THB-20μg/mL group,OGD+THB-40μg/mL group;Experimental group II group:normal Control group,LPS group,OGD+LPS group and OGD+LPS+THB-20μg/mL group.The relative expression of inflammation-related factors such as Caspase-1,ASC,IL-1βand IL-6 was detected by fluorescence quantitative PCR,and the expression of IL-1βand HO-1 was detected by Western Blot analysis.Results:The results of MTT assay showed that THB had no effect on the survival of EA.hy926 cells.The qPCR results of the experimental group I and II showed that the OGD model could play an anti-inflammatory protective role by down-regulating the expression of Caspase-1,ASC,IL-1βand IL-6 genes.The anti-inflammatory effect of THB on the OGD model was general,but it was found that THB also exerted an anti-inflammatory effect by down-regulating the expression of inflammatory genes by OGD+LPS group compared with OGD+LPS+THB-20μg/mL group.Western Blot analysis showed that the expression of IL-1βprotein was consistent with the results of qPCR.The expression of HO-1 protein as a protective factor was contrary to that of inflammatory factor fluorescence quantitative analysis.Conclusion:OGD model can improve the protective effect of cell stress.THB plays an anti-inflammatory role by down-regulating the expression of Caspase-1,ASC,IL-1βand IL-6 genes,up-regulating the expression of HO-1 protein and inhibiting the activation of NLRP3 inflammatory bodies.展开更多

Catalytic conversion of nitrate(NO_(3)^(-))pollutants into ammonia(NH_(3))offers a sustainable and promising route for both wastewater treatment and NH_(3)synthesis.Alkali cations are prevalent in nitrate solutions,bu...Catalytic conversion of nitrate(NO_(3)^(-))pollutants into ammonia(NH_(3))offers a sustainable and promising route for both wastewater treatment and NH_(3)synthesis.Alkali cations are prevalent in nitrate solutions,but their roles beyond charge balance in catalytic NO_(3)^(-)conversion have been generally ignored.Herein,we report the promotion effect of K^(+)cations in KNO_(3)solution for NO_(3)^(-)reduction over a TiO_(2)-supported Ni single-atom catalyst(Ni_(1)/TiO_(2)).For photocatalytic NO_(3)^(-)reduction reaction,Ni_(1)/TiO_(2)exhibited a 1.9-fold NH_(3)yield rate with nearly 100%selectivity in KNO_(3)solution relative to that in NaNO_(3)solution.Mechanistic studies reveal that the K^(+)cations from KNO_(3)gradually bonded with the surface of Ni_(1)/TiO_(2),in situ forming a K-O-Ni moiety during reaction,whereas the Na^(+)ions were unable to interact with the catalyst in NaNO_(3)solution.The charge accumulation on the Ni sites induced by the incorporation of K atom promoted the adsorption and activation of NO_(3)^(-).Furthermore,the K-O-Ni moiety facilitated the multiple proton-electron coupling of NO_(3)^(-)into NH_(3)by stabilizing the intermediates.展开更多
The clearance of apoptotic cell debris,containing professional phagocytosis and non-professional phagocytosis,is essential for maintaining the homeostasis of healthy tissues.Here,we discovered that endothelial cells c...The clearance of apoptotic cell debris,containing professional phagocytosis and non-professional phagocytosis,is essential for maintaining the homeostasis of healthy tissues.Here,we discovered that endothelial cells could engulf apoptotic cell debris in atherosclerotic plaque.Single-cell RNA sequencing(RNA-seq)has revealed a unique endothelial cell subpopulation in atherosclerosis,which was strongly associated with vascular injury-related pathways.Moreover,integrated analysis of three vascular injury-related RNA-seq datasets showed that the expression of scavenger receptor class B type 1(SR-B1)was up-regulated and specifically enriched in the phagocytosis pathway under vascular injury circumstances.Single-cell RNA-seq and bulk RNA-seq indicate that SR-B1 was highly expressed in a unique endothelial cell subpopulation of mouse aorta and strongly associated with the reorganization of cellular adherent junctions and cytoskeleton which were necessary for phagocytosis.Furthermore,SR-B1 was strongly required for endothelial cells to engulf apoptotic cell debris in atherosclerotic plaque of both mouse and human aorta.Overall,this study demonstrated that apoptotic cell debris could be engulfed by endothelial cells through SR-B1 and associated with the reorganization of cellular adherent junctions and cytoskeleton.展开更多
Developing bioactive materials for bone implants to enhance bone healing and bone growth has for years been the focus of clinical research.Barium titanate(BT)is an electroactive material that can generate electrical s...Developing bioactive materials for bone implants to enhance bone healing and bone growth has for years been the focus of clinical research.Barium titanate(BT)is an electroactive material that can generate electrical signals in response to applied mechanical forces.In this study,a BT piezoelectric ceramic coating was synthesized on the surface of a TC4 titanium alloy,forming a BT/TC4 material,and low-intensity pulsed ultrasound(LIPUS)was then applied as a mechanical stimulus.The combined effects on the biological responses of MC3T3-E1 cells were investigated.Results of scanning electron microscopy,energy-dispersive X-ray spectroscopy,and X-ray diffraction showed that an uniform nanospheres-shaped BT coating was formed on TC4 substrate.Piezoelectric behaviors were observed using piezoelectric force microscopy with the piezoelectric coefficient d_(33)of 0.42 pC/N.Electrochemical measures indicated that LIPUS-stimulated BT/TC4 materials could produce a microcurrent of approximately 10μA/cm^(2).In vitro,the greatest osteogenesis(cell adhesion,proliferation,and osteogenic differentiation)was found in MC3T3-E1 cells when BT/TC4 was stimulated using LIPUS.Furthermore,the intracellular calcium ion concentration increased in these cells,possibly because opening of the L-type calcium ion channels was promoted and expression of the Ca_(V)1.2 protein was increased.Therefore,the piezoelectric BT/TC4 material with LIPUS loading synergistically promoted osteogenesis,rending it a potential treatment for early stage formation of reliable bone-implant contact.展开更多

The Raman bands of morphine are assigned to certain molecule vibrations. The broad band in the long-wavelength region of the electronic absorption spectra of the sol with added adsorbent at certain concentrations has been explained in terms of the ag- gregation of the colloidal silver particles. The potential applications of SERS in quantitative measurement of the morphine samples are demonstrated. By using a proper Raman band of morphine, the detection limit of morphine in silver sol is found to be 1.5 ng/ml. The result suggests that it is of great significance to use SERS in illicit drug morphine inspection.展开更多
Highly efficient and durable water oxidation electrocatalysts are critically important in a wide range of clean energy technologies,including water electrolyzers and rechargeable metal-air batteries.Here,we report a n...Highly efficient and durable water oxidation electrocatalysts are critically important in a wide range of clean energy technologies,including water electrolyzers and rechargeable metal-air batteries.Here,we report a novel sonochemical approach to synthesize amorphous nickel-iron oxides/carbon nanohybrids with tunable compositions for the oxygen evolution reaction (OER).The sonochemically synthesized amorphous electrocatalysts with optimal composition exhibit a low overpotential of 290 mV at 10 mA&#183;cm-2 and a Tafel slope of 31 mV&#183;decade-1 in a 0.1 M KOH electrolyte,outperforming the benchmark RuO2 catalyst.Meanwhile,these nanohybrids are also highly stable and remain amorphous even after prolonged cycling.In addition to amorphism,sonochemistry endows as-prepared nickel-iron oxides/carbon nanohybrids with a simultaneously formed carbon scaffold and internal Ni(0),which can enhance the stability and activity for the OER.This work demonstrates that sonochemistry is a unique method for synthesizing amorphous metal oxides toward an efficient and durable OER.展开更多
Purpose:This review aims to explore the history,research hotspots,and emerging trends of drug-eluting stents(DES)in the last two decades from the perspective of structural and temporal dynamics.Methods:Publications on...Purpose:This review aims to explore the history,research hotspots,and emerging trends of drug-eluting stents(DES)in the last two decades from the perspective of structural and temporal dynamics.Methods:Publications on DES were retrieved from WoSCC.The bibliometric tools including CiteSpace and HistCite were used to identify the historical features,the evolution of active topics,and emerging trends on the DES field.Results:In the last 20 years,the field of DES is still in the hot phase and there is a wide range of extensive scientific collaborations.In addition,active topics emerge in different periods,as evidenced by a total of 41 disciplines,511 keywords,and 1377 papers with citation bursts.Keyword clustering anchored five emerging research subfields,namely#0 dual antiplatelet therapy,#3 drug-coated balloon,#4 bifurcation,5#rotational atherectomy,and 6#quantitative flow ratio.The keyword alluvial map shows that the most persistent research concepts in this field are thrombosis,restenosis,etc.,and the emerging keywords are paclitaxel eluting balloon,coated balloon,drug-eluting balloon,etc.There are 7 recent research subfields anchored by reference clustering,namely#2 dual antiplatelet therapy,#4 drug-coated balloon,#5 peripheral artery disease,#8 fractional flow reserve,#10 bioresorbable vascular scaffold,#13 intravascular ultrasound,#14 biodegradable polymer.Conclusion:The findings based on the bibliometric studies provide the current status and trends in DES research and may help researchers to identify hot topics and explore new research directions in this field.展开更多

Polycrystalline diamond compacts (PDC) were synthesized using diamond powder of average crystal size 3-20 μm by the Ni 70 Mn 25 Co 5 alloy infiltration technique at high temperature and high pressure (HPHT).The surfa...Polycrystalline diamond compacts (PDC) were synthesized using diamond powder of average crystal size 3-20 μm by the Ni 70 Mn 25 Co 5 alloy infiltration technique at high temperature and high pressure (HPHT).The surface residual stress of polycrystalline diamond (PCD) layer was measured using micro-Raman spectroscopy with hydrostatic stress model and X-ray diffraction (XRD).Measurements of the stress levels of PCDs show that the residual compressive stresses range from 0.12 to 0.22 GPa,which increase with the crystal size of diamond.Scanning electron microscopy (SEM) was used to observe the morphology of initial diamond grains and PCD cross-section.The results indicate that PCD has a dense and interlaced microstructure with diamond-diamond (D-D) direct bonding.And the smaller the crystal size of diamond,the better the growth of diamond direct bonding and the smaller the binder metal between diamond boundaries will be.展开更多

文摘D-D’-A type aza-borondipyrromethenes(aza-BODIPYs)were prepared by Suzuki cross-coupling reaction.Photothermal conversion efficiency of self-assemble aza-BODIPY-based nanoparticles(DA-azaBDP-NPs)with NIR-Ⅱ emission(λ_(em)=1065 nm)was 37.2%under near infrared(NIR)irradiation,and the outstanding cytotoxicity was triggered by coexistence of DA-azaBDP-NPs and the NIR irradiation,with the decrease of glioblastoma migration and the inhibition of glioblastoma proliferation.DA-azaBDP-NPs could promote glioblastoma autophagy and accelerate the process of cell death.The photothermal therapy(PTT)of DAazaBDP-NPs can effectively induce glioblastoma death by apoptosis under the NIR irradiation,which is highly promising to be applied in vivo experiments of brain.

文摘Objective:To explore the protective effect and possible mechanism of action of Zhuyu Annao pill in mice with intracerebral hemorrhage(ICH).Methods:Sixty mice were divided into the control group,hemorrhage group,drug-treated group(after hemorrhage),TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group(after hemorrhage) with 12 in each group.Model of autologous ICH was established in all groups.After drilling and 12 h of fasting,models in the control group hemorrhage group and TLR4-knockout hemorrhage group were all drenched with 10 mL/kg distilled water by intragastric administration.Models in the drug-treated group and TLR4-knockout hemorrhage + drugtreated group were drenched with 6.25 g/kg of Zhuyu Annao pill.All groups were treated for 7 d.Longa scoring method was used to measure the neurological defect scores and determine the brain water contents of all groups;ELISA was employed to detect the inflammatory factor interleukin(IL)-6,tumor necrosis factor- α(TNF- α) and IL-1β in brain tissues;and Western blot was applied to test the expression quantities of apoptotic protein Bax and anti-apoptotic protein Bcl-2 in brain tissues.Results:At day 3 and7,compared with the hemorrhage group,the neurological defect scores of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group decreased significantly(P<0.05) Compared with the hemorrhage group,the brain water contents of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group reduced significantly(P<0.05) Compared with the hemorrhage group,the inflammatory factor IL-6,TNF-α and IL-1β of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage + drug-treated group decreased significantly(P<0.05).Compared with the hemorrhage group,the expression of apoptotic protein Bax of the drug-treated group,TLR4-knockout hemorrhage group and TLR4-knockout hemorrhage+ drug-treated group decreased significantly and the expression of anti-apoptotic protein Bcl-2 increased significantly(P<0.05).Conclusions:Zhuyu Annao pill can alleviateencephaledema for mice with ICH and reduce inflammatory responsesandnerve cell apoptosis.TLR4 can mediate inflammatory injury induced by ICH.Thus,Zhuyu Annao pill can play a protective role for brains by decreasing the expression of TLR4.
文摘CS-CMC bipolar membrane was prepared and the cross-section photograph of CS-CMC BM was observed by SEM. FT-IR spectrum indicated that CS-CMC BM contained -N=CRH2^+ and -COO^- functional groups. The charge density of -N--CRH2^+ in CS membrane was about 14.13 mmol/g and the charge density of -COO in CMC membrane was about 9.01 mmol/g. The electrochemistry properties of CS-CMC BM were also studied. CS-CMC BM not only can effectively stop FeO4^2- from diffusing into the cathode chamber, but also plays an important role in the supply of OH" consumed during the electro-generated FeO4^2- process.

文摘Poly(vinylidene fluoride)(PVDF)/multi-walled carbon nanotube(MWCNT) nanocomposites were prepared by means of ultrasonic dispersion method. X-ray diffraction(XRD) results indicate that incorporating MWCNTs into PVDF caused the formation of β phase. A thermal annealing at 130 ℃ confirmed that the β phase was stable in the nanocomposites. Differential scanning calorimetry(DSC) results indicate that the melting temperature slightly increased while the heat of fusion markedly decreased with increasing MWCNT content. The tensile strength and modulus of PVDF were improved by loading the MWCNTs. The scanning electron microscopy(SEM) observations showed that MWCNTs were uniformly dispersed in the PVDF matrix and an interfacial adhesion between MWCNT and PVDF was achieved, which was responsible for the enhancement in the tensile strength and modulus of PVDE.

文摘The effect of H2O2 pretreatment on Cd tolerance and translocation of rice seedlings were studied using two rice cultivars (N07-6 and N07-63) differing in Cd tolerance. The contents of malondialdehyde (MDA), reduced glutathione (GSH), non-protein thiols (NPT), phytochelatins (PCs) and the activity of glutathione S-transferase (GST) were compared between the two cultivars exposed to various treatments. The results showed that 50 μmol/L Cd exposure significantly inhibited rice growth, enhanced the production of GSH, NPT, PCs and MDA, and increased the activity of GST, and there were significant differences between the two cultivars. More Cd was transported into the shoot of N07-6. The H2O2 pretreatment alleviated Cd toxicity by further increasing GSH, NPT and PCs contents, as well as the GST activity in roots. The increase degrees of these parameters in N07-63 were higher than those in N07-6, suggesting that the tolerance of N07-63 was enhanced more significantly than N07-6. Hydrogen peroxide reduced Cd translocation to rice shoot but affected the Cd content in root differently. From the above results, it may be speculated that there were remarkable differences in the Cd detoxification and response to the H2O2 pretreatment between the two cultivars.

文摘Background:Alzheimer’sdisease(AD)is a prevalent neurodegenerative disorder causing progressive dementia.Research suggests that microRNAs(miRNAs)could serve as biomarkers and therapeutic targets for AD.Reduced levels of miR-137 have been observed in the brains of AD patients,but its specific role and down stream mechanisms remain unclear.This study sought to examine the therapeutic potential of miR-137-5p agomir in alleviating cognitive dysfunction induced in AD models and explore its potential mechanisms.Methods:This study utilized bioinformatic analysis and a dual-l uciferase reporter assay to investigate the relationship between miR-137-5p and ubiquitin-specific peptidase 30(USP30).In vitro experiments were conducted using SH-SY5Y cells to assess the impact of miR-137-5p on Aβ1-42 neurotoxicity.In vivo experiments on AD mice evaluated the effects of miR-137-5p on cognition,Aβ1-42 deposition,Tau hyperphosphorylation,and neuronal apoptosis,as well as its influence on USP30 levels.Results:It was discovered that miR-137-5p mimics efficiently counteract Aβ1-42 neurotoxicity in SH-SY5Y cells,a protective effect that is negated by USP30 overexpression.In vivo experiments demonstrated that miR-137-5p enhances the cognition and mobility of AD mice,significantly reducing Aβ1-42 deposition,Tau hyperphosphorylation,and neuronal apoptosis within the hippocampus and cortex regions.Mechanistically,miR-137-5p significantly suppresses USP30 levels in mice,though USP30 overexpression partially buffers against miR-137-5p-i nduced AD symptom improvement.Conclusion:Our study proposes that miR-137-5p,by instigating the downregulation of USP30,has the potential to act as a novel and promising therapeutic target for AD.

文摘This study was conducted to investigate the effects of different mass concentrations of nitrobeneze on the growth, soluble sugar content, soluble protein content, chlorophyll a content and algal cell conductivity of Nitzschia hantzschia. The results showed that as the concentration of nitrobenzene increased, the growth of N. hantzschia was inhibited, and the algal culture liquids gradually changed from dark yellow to light yellow after 5 d of treatment;the soluble sugar content increased after 2 d;and the soluble protein content of the 100 mg/L nitrobenzene treatment group was 89.1% of the control group on day 1, which was the lowest value, and then showed a gradual upward trend. The low-mass concentration of nitrobenzene promoted the chlorophyll a content of algal cells, the medium and high mass concentrations had an inhibitory effect, and the chlorophyll a content of the 50 mg/L treatment gradually recovered after 3 d. The electrical conductivity of algal cells gradually increased with the increase of the mass concentration of nitrobenzene. The electrical conductivity gradually recovered after 3 d of the low mass concentration treatment, while the high mass concentration harmed the algae cells, causing N. hantzschia to gradually die.

文摘In this work, we designed a facile and promising strategy for fluorescence determination of S1 endonuclease activity and inhibition based on fluorescence resonance energy transfer(FRET) between positively Ag nanorods(AgNRs) and negatively-charged ROX-labeled sing-stranded DNA(ROX-ssDNA). In the absence of Sl endonuclease, the fluorescence signal of the ROX-ssDNA was efficiently quenched when the ROX-ssDNA was adsorbed on the surface of AgNRs via strong electrostatic interaction. However, upon addition of different concentration of Sl endonuclease, the fluorescence was gradually restored owing to the reduction of FRET efficiency caused by S1 endonuclease specific cleavage ROX-ssDNA into short fragments, which reduced the electrostatic interaction between AgNRs and short oligonucleotide fragment. This assay strategy exhibited a high sensitivity and excellent specificity for S1 endonuclease with a detection limit of 0.004 U/mL and a dynamic concentration range from 0.01 U/mL to 5.0 U/mL. In addition, the capabilities for screening of S1 endonuclease inhibitors and S1 endonuclease detection from complex biological matrixes were also verified.
文摘Objective: To investigate the law of age-related changes of insulin-like growth factor-1 (IGF-1) expression of rat thyroid cells cultured in vitro. Methods: Rat thyroid of different age (10, 45, 65, 100, 150 weeks) was isolated and thyrocytes cultured. Total RNA was extracted in different rat age group when thyroid cells had been cultured for two weeks. mRNA IGF-1 expression was measured with reverse-transcription polymerase chain reaction( RT-PCR) in each group and compared. Results: Quantity of total RNA in thyroid cells decreased with ageing when the rat thyroid cells had been cultured for 2 weeks. There is significant difference among groups (P<0.05). Expression of IGF-1 mRNA could be detected in thyroid cells of different age cultured in vitro. Quantity of IGF-1 mRNA expression by RT-PCR analysis increased from 10 to 45 weeks old, and then decreased with ageing. Conclusion: Rat thyroid cells from different age cultured in vitro can express IGF-1 mRNA. Quantity of total RNA in thyroid cells cultured in vitro decreased with aging. IGF-1 mRNA expression was correlated to age (r=0.401, P<0.05).

文摘The effects of light on the quality and related genes of Dendrobium officinale were summarized, and the main chemical components and pharmacological effects of D. officinale were analyzed. The biomass accumulation of D. officinale is related to light quality and light intensity. Monochromatic light quality can make polysaccharide accumulate, and mixed light quality can promote cell proliferation and component accumulation of D. officinale. Under different light intensity, the polysaccharide content of D. officinale is different, and it changes with the change of light intensity. When a certain intensity is exceeded, the polysaccharide content decreases with the increase of light intensity. The chemical components of D. officinale are polysaccharides, alkaloids, amino acids, volatile components, etc. The analysis of the growth and components of D. officinale provides reference and basis for improving the quality and chemical components of D. officinale and promotes the research and development of related products of D. officinale.

文摘Objective:To investigate the effects and mechanisms of tetrahydroberberine(THB)on vascular endothelial cell injury induced by oxygen glucose deprivation(OGD)and LPS-induced cell injury model.Methods:MTT assay was used to detect the effects of THB final concentration on the survival rate of EA.hy926 cells,which were 0μg/mL,2.5μg/mL,5μg/mL,10μg/mL,20μg/mL and 40μg/mL,respectively.Group I of the experiment:normal control group,OGD group,OGD+THB-10μg/mL group,OGD+THB-20μg/mL group,OGD+THB-40μg/mL group;Experimental group II group:normal Control group,LPS group,OGD+LPS group and OGD+LPS+THB-20μg/mL group.The relative expression of inflammation-related factors such as Caspase-1,ASC,IL-1βand IL-6 was detected by fluorescence quantitative PCR,and the expression of IL-1βand HO-1 was detected by Western Blot analysis.Results:The results of MTT assay showed that THB had no effect on the survival of EA.hy926 cells.The qPCR results of the experimental group I and II showed that the OGD model could play an anti-inflammatory protective role by down-regulating the expression of Caspase-1,ASC,IL-1βand IL-6 genes.The anti-inflammatory effect of THB on the OGD model was general,but it was found that THB also exerted an anti-inflammatory effect by down-regulating the expression of inflammatory genes by OGD+LPS group compared with OGD+LPS+THB-20μg/mL group.Western Blot analysis showed that the expression of IL-1βprotein was consistent with the results of qPCR.The expression of HO-1 protein as a protective factor was contrary to that of inflammatory factor fluorescence quantitative analysis.Conclusion:OGD model can improve the protective effect of cell stress.THB plays an anti-inflammatory role by down-regulating the expression of Caspase-1,ASC,IL-1βand IL-6 genes,up-regulating the expression of HO-1 protein and inhibiting the activation of NLRP3 inflammatory bodies.

文摘Catalytic conversion of nitrate(NO_(3)^(-))pollutants into ammonia(NH_(3))offers a sustainable and promising route for both wastewater treatment and NH_(3)synthesis.Alkali cations are prevalent in nitrate solutions,but their roles beyond charge balance in catalytic NO_(3)^(-)conversion have been generally ignored.Herein,we report the promotion effect of K^(+)cations in KNO_(3)solution for NO_(3)^(-)reduction over a TiO_(2)-supported Ni single-atom catalyst(Ni_(1)/TiO_(2)).For photocatalytic NO_(3)^(-)reduction reaction,Ni_(1)/TiO_(2)exhibited a 1.9-fold NH_(3)yield rate with nearly 100%selectivity in KNO_(3)solution relative to that in NaNO_(3)solution.Mechanistic studies reveal that the K^(+)cations from KNO_(3)gradually bonded with the surface of Ni_(1)/TiO_(2),in situ forming a K-O-Ni moiety during reaction,whereas the Na^(+)ions were unable to interact with the catalyst in NaNO_(3)solution.The charge accumulation on the Ni sites induced by the incorporation of K atom promoted the adsorption and activation of NO_(3)^(-).Furthermore,the K-O-Ni moiety facilitated the multiple proton-electron coupling of NO_(3)^(-)into NH_(3)by stabilizing the intermediates.

文摘The clearance of apoptotic cell debris,containing professional phagocytosis and non-professional phagocytosis,is essential for maintaining the homeostasis of healthy tissues.Here,we discovered that endothelial cells could engulf apoptotic cell debris in atherosclerotic plaque.Single-cell RNA sequencing(RNA-seq)has revealed a unique endothelial cell subpopulation in atherosclerosis,which was strongly associated with vascular injury-related pathways.Moreover,integrated analysis of three vascular injury-related RNA-seq datasets showed that the expression of scavenger receptor class B type 1(SR-B1)was up-regulated and specifically enriched in the phagocytosis pathway under vascular injury circumstances.Single-cell RNA-seq and bulk RNA-seq indicate that SR-B1 was highly expressed in a unique endothelial cell subpopulation of mouse aorta and strongly associated with the reorganization of cellular adherent junctions and cytoskeleton which were necessary for phagocytosis.Furthermore,SR-B1 was strongly required for endothelial cells to engulf apoptotic cell debris in atherosclerotic plaque of both mouse and human aorta.Overall,this study demonstrated that apoptotic cell debris could be engulfed by endothelial cells through SR-B1 and associated with the reorganization of cellular adherent junctions and cytoskeleton.

文摘Developing bioactive materials for bone implants to enhance bone healing and bone growth has for years been the focus of clinical research.Barium titanate(BT)is an electroactive material that can generate electrical signals in response to applied mechanical forces.In this study,a BT piezoelectric ceramic coating was synthesized on the surface of a TC4 titanium alloy,forming a BT/TC4 material,and low-intensity pulsed ultrasound(LIPUS)was then applied as a mechanical stimulus.The combined effects on the biological responses of MC3T3-E1 cells were investigated.Results of scanning electron microscopy,energy-dispersive X-ray spectroscopy,and X-ray diffraction showed that an uniform nanospheres-shaped BT coating was formed on TC4 substrate.Piezoelectric behaviors were observed using piezoelectric force microscopy with the piezoelectric coefficient d_(33)of 0.42 pC/N.Electrochemical measures indicated that LIPUS-stimulated BT/TC4 materials could produce a microcurrent of approximately 10μA/cm^(2).In vitro,the greatest osteogenesis(cell adhesion,proliferation,and osteogenic differentiation)was found in MC3T3-E1 cells when BT/TC4 was stimulated using LIPUS.Furthermore,the intracellular calcium ion concentration increased in these cells,possibly because opening of the L-type calcium ion channels was promoted and expression of the Ca_(V)1.2 protein was increased.Therefore,the piezoelectric BT/TC4 material with LIPUS loading synergistically promoted osteogenesis,rending it a potential treatment for early stage formation of reliable bone-implant contact.

文摘We report the surface-enhanced Raman (SERS) spectra of morphine in silver colloid, and study the silver colloid enhanced effects on the Raman scattering of morphine. The Raman bands of morphine are assigned to certain molecule vibrations. The broad band in the long-wavelength region of the electronic absorption spectra of the sol with added adsorbent at certain concentrations has been explained in terms of the ag- gregation of the colloidal silver particles. The potential applications of SERS in quantitative measurement of the morphine samples are demonstrated. By using a proper Raman band of morphine, the detection limit of morphine in silver sol is found to be 1.5 ng/ml. The result suggests that it is of great significance to use SERS in illicit drug morphine inspection.

文摘Highly efficient and durable water oxidation electrocatalysts are critically important in a wide range of clean energy technologies,including water electrolyzers and rechargeable metal-air batteries.Here,we report a novel sonochemical approach to synthesize amorphous nickel-iron oxides/carbon nanohybrids with tunable compositions for the oxygen evolution reaction (OER).The sonochemically synthesized amorphous electrocatalysts with optimal composition exhibit a low overpotential of 290 mV at 10 mA&#183;cm-2 and a Tafel slope of 31 mV&#183;decade-1 in a 0.1 M KOH electrolyte,outperforming the benchmark RuO2 catalyst.Meanwhile,these nanohybrids are also highly stable and remain amorphous even after prolonged cycling.In addition to amorphism,sonochemistry endows as-prepared nickel-iron oxides/carbon nanohybrids with a simultaneously formed carbon scaffold and internal Ni(0),which can enhance the stability and activity for the OER.This work demonstrates that sonochemistry is a unique method for synthesizing amorphous metal oxides toward an efficient and durable OER.

文摘Purpose:This review aims to explore the history,research hotspots,and emerging trends of drug-eluting stents(DES)in the last two decades from the perspective of structural and temporal dynamics.Methods:Publications on DES were retrieved from WoSCC.The bibliometric tools including CiteSpace and HistCite were used to identify the historical features,the evolution of active topics,and emerging trends on the DES field.Results:In the last 20 years,the field of DES is still in the hot phase and there is a wide range of extensive scientific collaborations.In addition,active topics emerge in different periods,as evidenced by a total of 41 disciplines,511 keywords,and 1377 papers with citation bursts.Keyword clustering anchored five emerging research subfields,namely#0 dual antiplatelet therapy,#3 drug-coated balloon,#4 bifurcation,5#rotational atherectomy,and 6#quantitative flow ratio.The keyword alluvial map shows that the most persistent research concepts in this field are thrombosis,restenosis,etc.,and the emerging keywords are paclitaxel eluting balloon,coated balloon,drug-eluting balloon,etc.There are 7 recent research subfields anchored by reference clustering,namely#2 dual antiplatelet therapy,#4 drug-coated balloon,#5 peripheral artery disease,#8 fractional flow reserve,#10 bioresorbable vascular scaffold,#13 intravascular ultrasound,#14 biodegradable polymer.Conclusion:The findings based on the bibliometric studies provide the current status and trends in DES research and may help researchers to identify hot topics and explore new research directions in this field.

文摘Exosomes are membrane vesicles that exist in pathological or normal cells and are considered to be specifically secreted,contain complex RNAs and proteins,participate in intercellular communication functions,and have functions in organismal immune response,antigen presentation,cell migration,cell differentiation,and tumor invasion.In recent years,with the explosion of the development of minimally invasive diagnostics,the research of exosomes has been rapidly developed,and the number of articles published in this field has increased dramatically,and many articles summarize the published papers in a review-type analysis.In this paper,we use bibliometric analysis combined with theoretical analysis and pathfinding network scaling to visualize data and objectively count the development process,current research,and development trend of the corresponding field by analyzing the literature of the specified field.This paper presents the data visualization by analyzing the literature of the specified field to objectively count the development process,current research and development trend of the corresponding field.This paper presents a retrospective analysis and review of the articles on exosomes in the last 20 years(2001–2021)based on the bibliometric analysis method of CiteSpace software,while presenting the problems of current research and discussing the challenges encountered in the future,hoping to provide new ideas for the development of research in this field.

文摘Polycrystalline diamond compacts (PDC) were synthesized using diamond powder of average crystal size 3-20 μm by the Ni 70 Mn 25 Co 5 alloy infiltration technique at high temperature and high pressure (HPHT).The surface residual stress of polycrystalline diamond (PCD) layer was measured using micro-Raman spectroscopy with hydrostatic stress model and X-ray diffraction (XRD).Measurements of the stress levels of PCDs show that the residual compressive stresses range from 0.12 to 0.22 GPa,which increase with the crystal size of diamond.Scanning electron microscopy (SEM) was used to observe the morphology of initial diamond grains and PCD cross-section.The results indicate that PCD has a dense and interlaced microstructure with diamond-diamond (D-D) direct bonding.And the smaller the crystal size of diamond,the better the growth of diamond direct bonding and the smaller the binder metal between diamond boundaries will be.
