Three suggestions were made for further discoveries of new actinomycetes. First, natural habitats such as old growth forests, and extreme environments, should be given sampling priority; second, culturing procedures need to be constantly improved to mimic natural habitats; third, less than 98.5% similarity of 16S rRNA sequence to that of a known species could be used as an indicator of a new species. 展开更多

The Rehai Geothermal Field, located in Tengchong County, in central-western Yunnan Prov- ince, is the largest and most intensively studied geothermal field in China. A wide physicochemical diversity of springs (ambient to -97 ℃; pH from 〈1.8 to≥9.3) provides a multitude of niches for extremophilic microorganisms. A variety of studies have focused on the cultivation, identification, basic physiology, taxonomy, and biotechnological potential of thermophilic microorganisms from Rehai. Ther- mophilic bacteria isolated from Rehai belong to the phyla Firmicutes and Deinococcus-Thermus. Firmicutes include neutrophilic or alkaliphilic Anoxybacillus, Bacillus, Caldalkalibacillus, Caldanaerobacter, Laceyella, and Geobacillus, as well as thermoacidophilic Alicyclobacillus and Sulfobacillus. Isolates from the Deinococcus-Thermus phylum include several Meiothermus and Thermus species. Many of these bacteria synthesize thermostable polymer-degrading enzymes that may be useful for biotech- nology. The thermoacidophilic archaea Acidianus, Metallosphaera, and Sulfolobus have also been isolated and studied. A few studies have reported the isolation of thermophilic viruses belonging to Siphoviridae (TTSP4 and TTSP10) and Fuselloviridae (STSV1) infecting Thermus spp. and Sulfolobus spp., respectively. More recently, cultivation-independent studies using 16S rRNA gene sequences, shotgun metagenomics, or "'functional gene" sequences have revealed a mtlch broader diversity of micro- organisms than represented in culture. Studies of the gene and mRNA encoding 113e large subunit of the ammonia monooxygenase (amoA) of ammonia-oxidizing Archaea (AOA) and the tetraether lipid cre- narchaeol, a potential hiomarker for AOA, suggest a wide diversity, but possibly low abundance, of ther- mophilic AOA in Rehai. Finally, we introduce the Tengchong Partnerships in International Research and Education (P1RE) project, an international collaboration between Chinese and U.S. scientists with the goal of promoting international and interdisciplinary cooperation to gain a more holistic and gh〉bal view of life in te^estrial geothermal springs. 展开更多

Anti-aging research has become a popular scientific field with the increasing prominence of population aging.Rare ginsenoside Compound K(CK)has attracted widespread attention as an emerging anti-aging active ingredient.The anti-aging effect of ginsenosides is considered to be one of the important roles of ginsenosides,and Compound K,as the main deglycosylated metabolite of ginsenosides,has a comprehensive anti-aging effect as a highly active ingredient obtained by transformation under the action of microbiota.Recent studies have shown that ginsenosides have anti-photo-oxidation,anti-skin aging,free radical scavenging and immunostimulatory effects,which can effectively prevent skin photoaging.With the progress of modern natural medicine extraction technology and the deepening of the research on the anti-skin aging of ginsenosides'high active ingredients,it will promote the development and application of natural product protective skin photoaging preparations.The rare ginsenoside Compound K plays an important role in the improvement of skin health and anti-aging,which is mainly realized by increasing the activity of antioxidant enzymes,inducing the expression of related genes,reducing the content of oxidative damage substances,regulating the immune system,and influencing the expression of cell-cycle regulators and aging genes.A more comprehensive and in-depth study of the molecular mechanism of the anti-aging effect of rare ginsenoside Compound K will be one of the focuses of future research. 展开更多

Although the efficiency of poly(ethylene terephthalate)(PET)degradation has been successfully improved by depolymerase engineering,mostly by using Goodfellow-PET(gf-PET)as a substrate,efforts to degrade unpretreated PET materials with high crystallinity remain insufficient.Here,we endeavored to improve the degradation capability of a WCCG mutant of leaf-branch compost cutinase(LCC)on a unpretreated PET substrate(crystallinity>40%)by employing iterative saturation mutagenesis.Using this method,we developed a high-throughput screening strategy appropriate for unpretreated substrates.Through extensive screening of residues around the substrate-binding groove,two variants,WCCG-sup1 and WCCG-sup2,showed good depolymerization capabilities with both high-(42%)and low-crystallinity(9%)substrates.The WCCG-sup1 variant completely depolymerized a commercial unpretreated PET product in 36 h at 72℃.In addition to enzyme thermostability and catalytic efficiency,the adsorption of enzymes onto substrates plays an important role in PET degradation.This study provides valuable insights into the structure-function relationship of LCC. 展开更多

Background:The forest musk deer,a rare fauna species found in China,is famous for its musk secretion which is used in selected Traditional Chinese medicines.However,over-hunting has led to musk deer becoming an endangered species,and their survival is also greatly challenged by various high incidence and high mortality respiratory and intestinal diseases such as septic pneumonia and enteritis.Accumulating evidence has demonstrated that Akkermannia muciniphila(AKK)is a promising probiotic,and we wondered whether AKK could be used as a food additive in animal breeding pro-grammes to help prevent intestinal diseases.Methods:We isolated one AKK strain from musk deer feces(AKK-D)using an im-proved enrichment medium combined with real-time PCR.After confirmation by 16S rRNA gene sequencing,a series of in vitro tests was conducted to evaluate the probiotic effects of AKK-D by assessing its reproductive capability,simulated gas-trointestinal fluid tolerance,acid and bile salt resistance,self-aggregation ability,hy-drophobicity,antibiotic sensitivity,hemolysis,harmful metabolite production,biofilm formation ability,and bacterial adhesion to gastrointestinal mucosa.Results:The AKK-D strain has a probiotic function similar to that of the standard strain in humans(AKK-H).An in vivo study found that AKK-D significantly amelio-rated symptoms in the enterotoxigenic Escherichia coli(ETEC)-induced murine diar-rhea model.AKK-D improved organ damage,inhibited inflammatory responses,and improved intestinal barrier permeability.Additionally,AKK-D promoted the reconsti-tution and maintenance of the homeostasis of gut microflora,as indicated by the fact that AKK-D-treated mice showed a decrease in Bacteroidetes and an increase in the proportion of other beneficial bacteria like Muribaculaceae,Muribaculum,and unclas-sified f_Lachnospiaceae compared with the diarrhea model mice.Conclusion:Taken together,our data show that this novel AKK-D strain might be a potential probiotic for use in musk deer breeding,although further extensive system-atic research is still needed. 展开更多

Suicidality is a complex phenomenon influenced by genetic,environmental,and epigenetic factors.Current tools to estimate suicide risk are insufficient,and there is an increasing need for reliable biomarkers to complement clinical approaches.Growing evidence suggests that immune system dysregulation contributes to the pathophysiology of psychiatric disorders and suicidal behavior.Epigenetic mechanisms,including DNA methylation,histone modifications,and non-coding RNAs,regulate gene expression and may act as a bridge between environmental stressors and(neuro)inflammatory responses.In this review,we examine the evidence of peripheral and central inflammation in suicide completers and individuals with suicidal behavior.Next,we review current knowledge from various studies on suicide-associated epigenetic alterations.Furthermore,we evaluate the mechanisms by which early life adversity and chronic stress contribute to suicide diathesis,focusing on their association with epigenetic modifications and inflammatory pathways.We also examine future prospects and limitations of immunology-related biomarkers and the possibilities of therapeutic interventions targeting the immune system and epigenetic regulation.While challenging,research on epigenetic and immune alterations in suicidality shows promise for identifying suicide risk subtypes and advancing personalized psychiatry. 展开更多

Adaptation of lentils to high-temperature stress at phytopathogenic infection and application of molybdenum nanoparticles

The study of the impact of high-temperature stress on the plant-phytopathogen system,including using nanoparticles,is relevant in connection with climate changes.Therefore,our work was aimed at studying the impact of high-temperature stress on the physiological,biochemical,and growth processes of lentil plants under the conditions of pre-sowing treatment of seeds with Mo nanocitrates(Mo Ncit)and artificial infection with phytopathogenic microorganisms.Under high-temperature stress at 32-34°C(humidity 45 and 40%)in field conditions,a decrease in the photochemical activity of leaves of intact lentil plants was observed,due to the acclimatization of the photosynthetic apparatus.Infection with phytopathogenic phytoplasmas and bacteria disrupted the balance of free radicals in leaf tissues,because of which the activity of antioxidant enzymes SOD,POD,and phenolic compounds increased.This was likely the main reason for the disruption of the adaptation of the photosynthetic apparatus of plants to high-temperature stress under artificial infection with phytopathogenic phytoplasmas and bacteria,which destroyed pigment-protein complexes of PS II.As a result,the chlorophyll content in the leaves decreased.At the same time,physiological and biochemical changes in metabolism at presowing seed treatment with Mo Ncit compared to the control,caused an increase in the productivity of lentil plants in natural conditions under the combined action of various factors during the growing season:temperature fluctuations,air humidity,infection with phytopathogenic microorganisms.Therefore,infection with phytopathogenic microorganisms worsened the growth processes of plants,including due to a decrease in the adaptation of the photosynthetic apparatus to high-temperature stress.It is worth emphasizing that pre-treatment with molybdenum nanocitrates had a growth-stimulating and,to some extent,adaptive effect on plants and increased tolerance to phytopathogens. 展开更多

Several Trichoderma species serve as biocontrol agents in agriculture through their phytopathogen growth inhibition capabilities.However,the antagonistic mechanism of certain strains primarily operates through direct action.This study aims to explore an effective strain with comprehensive capabilities and elucidate its practical viability and action mechanism.Trichoderma gamsii strain TC959,exhibiting robust antagonistic and plant growth-promoting properties,was identified.The strain directly inhibits plant pathogen through the production of secondary metabolites,siderophores,and chitinase/xylanase,while promotes plant growth via indole-3-acetic acid/gibberellin release.Additionally,the strain activates induced systemic resistance by enhancing the chlorophyll a/b ratio and jasmonic acid content in pepper seedlings through root colonization,leading to elevated defense-related gene expression,antioxidant enzyme activity,and indole-3-acetic acid/gibberellin production.These mechanisms collectively enhance disease resistance and promote plant growth.Moreover,TC959 demonstrates superior resistance to oxidation and chemical fungicides,facilitating strain viability maintenance and ensuring healthy pepper seedling development.The study concludes that strain TC959 exhibits significant biocontrol potential and comprehensive functions against pepper damping-off disease,warranting further practical applications. 展开更多

Computational redesign of a thermostable MHET hydrolase and its role as an endo-PETase in promoting PET depolymerization

Biotechnological strategies for plastic depolymerization and recycling have emerged as transformative approaches to combat the global plastic pollution crisis,aligning with the principles of a sustainable and circular economy.Despite advances in engineering PET hydrolases,the degradation process is frequently compromised by product inhibition and the heterogeneity of final products,thereby obstructing subsequent PET recondensation and impeding the synthesis of high-value derivatives.In this work,we utilized previously devised computational strategies to redesign a thermostable DuraMHETase,achieving an apparent melting temperature of 72℃ in complex with MHET and a 6-fold higher in total turnover number(TTN)toward MHET than the wild-type enzyme at 60℃.The fused enzyme system composed of DuraMHETase and TurboPETase demonstrated higher efficiency than other PET hydrolases and the separated dual enzyme systems.Furthermore,we identified both exo-and endo-PETase activities in DuraMHETase,whereas the endo-activity was previously unobserved at ambient temperatures.These results expand the functional scope of MHETase beyond mere intermediate hydrolysis,and may provide guidance for the development of more synergistic approaches to plastic biodepolymerization and recycling. 展开更多

Background Rumen microorganisms are key regulators of ruminant growth and production performance.Identifying probiotic candidates through microbial culturomics presents a promising strategy for improving ruminant production performance.Our previous study identified significant differences in rumen microbial communities of Holstein calves with varying average daily gain(ADG).This study aims to identify a target strain based on the findings from multi-omics analysis and literature review,isolating and evaluating the target microbial strains from both the rumen and hindgut contents for their probiotic potential.Results Parabacteroides distasonis,a strain closely associated with ADG,was successfully isolated from calf rumen content cultured with Fastidious Anaerobe Agar(FAA)medium and named Parabacteroides distasonis F4.Wholegenome sequencing and pan-genome analysis showed that P.distasonis F4 possesses a core functional potential for carbohydrate and amino acid metabolism,with the ability to produce propionate,acetate,and lactate.The results of targeted and untargeted metabolomics further validated the organic acid production and metabolic pathways of P.distasonis F4.An in vitro simulated rumen fermentation test showed that supplementation with P.distasonis F4 significantly altered rumen microbial community structure and increased the molar proportions of propionate and butyrate in the rumen.Furthermore,an in vivo study demonstrated that dietary supplementation with P.distasonis F4 significantly increased the ADG of pre-weaning calves.Conclusions This study represents the first isolation of P.distasonis F4 from rumen,highlighting its potential as a probiotic strain for improving rumen development and growth performance in ruminants. 展开更多

Influenza A virus(IAV)remains a global public health concern,causing influenza-like illness and severe respiratory tract infections.Two major subtypes,A/pdm09 H1N1 and A/H3N2,circulate globally,and their epidemics are influenced by multiple factors,especially during the COVID-19 pandemic.Based on data from the National Influenza Surveillance Program in China,we analyzed the epidemiological and genomic data in Tianjin collected from 2017 to 2025.A total of 77,473 throat swabs were collected,of which 9144 were IAV-positive.The A/pdm09 H1N1 and A/H3N2 lineages exhibited distinct epidemics across different influenza seasons,with a decline in cases observed during the COVID-19 pandemic.We sequenced the genomes of 128 A/pdm09 H1N1 and 113 A/H3N2 clinical isolates and characterized their temporal evolution and genetic diversity using time-scaled phylogenetic analysis.Additionally,we conducted a genetic risk evaluation of the hemagglutinin and neuraminidase segments,identifying key amino acid residues associated with viral adaptation,transmissibility,virulence,and drug resistance.Moreover,no antigenic variants were found in clinical isolates during the recent influenza seasons,though reduced sensitivity to oseltamivir and zanamivir was observed in individual strains.Our surveillance highlights the epidemiology and evolution of IAV before and after the COVID-19 pandemic in Tianjin. 展开更多

Siraitia grosvenorii(SG),a traditional edible medicine in China,has shown potential application in health foods due to multiple bioactivities.However,few comparative studies have investigated the effect of daily intake of SG products by traditional drinking way on alleviating metabolic disorders caused by long-term high fat and high sugar diet(HF-HSD).This study investigated the effects of 2 commercial SG products(SG juice containing comprehensive compositions and mogrosides extract containing high content of mogroside V)supplement on fat content,lipid accumulation,colon barrier and gut microbiota in a 38-week HF-HSD mice trial.Results showed that the SG intervention could decrease the weight gain and body fat content,alleviated lipid accumulation,low-grade inflammation and colon barrier injury.SG juice intake was found to increase the antioxidant enzyme activities of serum glutathione peroxidase and liver superoxide dismutase.16S r RNA analysis found that SG could restore the gut microbiota with downregulation of harmful bacteria increased by HF-HSD and enhancement of the abundances of beneficial genus.In particular,SG juice significantly enriched taxonomic family Prevotellaceae and genus Alloprevotella.These results indicated that SG might be a functional additive to prevent disease risk caused by excess fat and sugar diet,and active ingredients resulting to the discrepancy of performance especially on gut microbiota are worthy of in-depth investigation. 展开更多

Conjunctival microbiota variations in a subset of middleaged and elderly individuals from Beijing,China

AIM:To isolate and identify the conjunctival microbiota of cataract patients and analyze the associated influencing factors.METHODS:This study recruited 216 participants(216 eyes)from April 2022 to July 2022.Under the condition of no antibiotic use prior to cataract surgery,sterile swabs were used to collect samples from the lower conjunctival sac.Bacterial cultures were then conducted,followed by species identification through 16S rDNA gene sequencing.Clinical factors associated with positive or negative bacterial isolation rates were analyzed,including age,gender,meibomian gland dysfunction(MGD),history of hypertension,history of diabetes,history of cancer,history of infectious diseases and the habit of wearing masks.RESULTS:Among the 216 eyes,78 eyes yielded isolates,with an isolation rate of 36.11%,detecting a total of 122 strains.Gram-positive rods accounted for 49.18%(60 strains),gram-positive cocci accounted for 45.08%(55 strains),gram-negative bacteria accounted for 4.92%(6 strains),and fungi accounted for 0.82%(1 strain).This study found that the most abundant genera in the conjunctival sac were Corynebacterium(42.62%),Staphylococcus(31.15%),Micrococcus(9.84%),Acinetobacter(4.10%),and Bacillus(3.28%).Furthermore,age(P=0.006),gender(P=0.039),diabetes(P=0.003),history of infectious diseases(P=0.02),and duration of mask replacement(P<0.001)were important factors influencing the positive bacterial culture of the conjunctival microbiota.Although hypertensive patients exhibited a higher isolation rate of conjunctival bacteria,it did not reach statistical significance,and the history of cancer did not affect the isolation rate of the conjunctival microbial community in cataract patients before surgery.CONCLUSION:Potential changes are observed in the conjunctival microbiota among a sample of middleaged and elderly individuals from Beijing,China.Notably,an increased isolation rate of Corynebacterium and Micrococcus is detected,suggesting a possible change in the microbial balance that requires further investigation and attention from the ophthalmological community.Advanced age,female gender,MGD,diabetes,a recent history of infectious diseases,and inadequate mask-wearing habits are potentially significant factors associated with the conjunctival microbiota.These factors should be considered in the development of strategies to prevent perioperative infections in cataract surgery patients. 展开更多

Hypersaline mariculture wastewater necessitates treatment prior to its discharge into marine environments.Algal-mycelial pellets(AMPs),known for their cost-effectiveness,energy efficiency and sustainability,have not been previously explored for their flocculation and pollutant removal capabilities in hyperhaline conditions.This work employed an orthogonal test design to investigate the effects of nine factors at three levels on the treatment efficiency of mariculture wastewater using Chlorella sp.TNBR1 and Aspergillus niger AMPs.The comprehensive optimal conditions for achieving the highest flocculation efficiency and pollutant removal are determined to be a temperature of 30℃,light intensity of 6000 lux,a 12:0 light-dark cycle,an initial pH of 6,amicroalgal density of 11.25×10^(6)cell/mL,microalgal growth phase at the early logarithmic stage,a fungal spore density of 9.00×10^(5)spore/mL and a fungal pellet phase of 60 h.Under such conditions,the treatment of nonsterile actual mariculture wastewater with Chlorella sp.TNBR1 and Aspergillus niger AMPs results in a 93.35%±7.20%reduction in chemical oxygen demand(COD),92.83%±7.29%reduction in total nitrogen(TN),100%removal of total phosphorus(TP),and a flocculation efficiency of 69.21%±5.36%.These findings confirm that AMPs are a viable solution for effectively treating COD,TN and TP in real hypersaline mariculture wastewater,while also facilitating the flocculation and harvesting of microalgae. 展开更多

The bioactivity-guided isolation of potentially active natural products has been widely utilized in pharmaceutical discovery.In this study,by screening fungal extracts against coxsackievirus B3(CVB3),three new aspochalasins,templichalasins A-C(1-3),along with six known aspochalasins(4-9)were isolated from an active extract derived from the endophytic fungus Aspergillus templicola LHWf045.Compound 1 features a unique 5/6/5/7/5 pentacyclic ring system,while compounds 2 and 3 possess unusual 5/6/6/7 tetracyclic skeletons.Their structures were characterized through extensive spectroscopic analyses,electronic circular dichroism(ECD)calculations,and single-crystal X-ray diffraction analysis.Additionally,we demonstrated that compound 4 can be readily converted into compounds 1-3 under mild acidic conditions and proposed a plausible mechanism for this conversion.Bioactivity evaluation of compounds 1-9 against CVB3 revealed the inhibitory effects of all compounds against the virus.Notably,compound 9 exhibited superior antiviral activity,surpassing the commercial drug ribavirin in selectivity index(SI)value. 展开更多

Human adenovirus type 108(HAdV-108)has been detected in multiple countries,including China,and is associated with severe acute respiratory infection(ARI)in children,with reported fatalities.However,studies on HAdV-108 remain limited.This study aimed to investigate the clinical and genetic characteristics of HAdV-108 in ARI children in China.From 2014 to 2024,6720 respiratory samples were collected from hospitalized children with ARI at ten hospitals across northern and southern China,of which 505(7.51%)tested positive for HAdV.The whole-genome and three major capsid protein genes were amplified and sequenced for bioinformatics analysis,which revealed that among 317 HAdV-isolated samples,21(6.62%)were identified as HAdV-108,ranking third after HAdV-114 and HAdV-7.Clinical analysis of HAdV-108-positive cases showed that the main manifestations were cough and fever.Seven children had gastrointestinal symptoms,and two children without underlying diseases were diagnosed with severe pneumonia.Phylogenetic analysis of wholegenome sequences revealed distinct predominant epidemic branches between domestic and international strains,with one strain obtained in this study forming an independent branch.Hexon protein exhibited the fastest evolution rate,lowest identity,and greatest amino acid variability,while fiber protein displayed the slowest evolution rate,highest identity,and greatest conservation and stability.Compared with the earliest reported HAdV-108 strain,three amino acid deletions were identified in the RGD loop region of penton base protein,resulting in potential structural change.Recombination analysis identified five distinct recombination patterns.In vitro experiments demonstrated that HAdV-108 had proliferation capacity comparable to other species C adenoviruses.In summary,HAdV-108 has persistently circulated in China,causing severe ARIs and concurrent gastrointestinal manifestations.Cluster3 was the predominant epidemic branch in China.HAdV-108 exhibited significant intratype genetic variation,with random and diverse recombination events. 展开更多

Metabolic syndrome characterized by obesity, hyperglycemia and liver steatosis is becoming prevalent all over the world. Herein, a water insoluble polysaccharide(WIP) was isolated and identified from the sclerotium of Poria cocos, a widely used Traditional Chinese Medicine. WIP was confirmed to be a(1-3)-β-D-glucan with an average Mw of 4.486 × 10~6 Da by NMR and SEC-RI-MALLS analyses. Furthermore, oral treatment with WIP from P. cocos significantly improved glucose and lipid metabolism and alleviated hepatic steatosis in ob/ob mice. 16 S DNA sequencing analysis of cecum content from WIP-treated mice indicated the increase of butyrate-producing bacteria Lachnospiracea, Clostridium. It was also observed that WIP treatment elevated the level of butyrate in gut, improved the gut mucosal integrity and activated the intestinal PPAR-γ pathway. Fecal transplantation experiments definitely confirmed the causative role of gut microbiota in mediating the benefits of WIP. It is the first report that the water insoluble polysaccharide from the sclerotium of P. cocos modulates gut microbiota to improve hyperglycemia and hyperlipidemia. Thereby, WIP from P. cocos, as a prebiotic, has the potential for the prevention or cure of metabolic diseases and may elucidate new mechanism for the efficacies of this traditional herbal medicine on the regulation of lipid and glucose metabolism. 展开更多

A psychrophilic aerobic denitrifying bacterium,strain S1-1,was isolated from a biological aerated filter conducted for treatment of recirculating water in a marine aquaculture system.Strain S1-1 was preliminarily identified as Psychrobacter sp.based on the analysis of its 16S rRNA gene sequence,which showed 100% sequence similarity to that of Psychrobacter sp.TSBY-70.Strain S1-1 grew well either in high nitrate or high nitrite conditions with a removal of 100% nitrate or 63.50% nitrite,and the total nitrogen removal rates could reach to 46.48% and 31.89%,respectively.The results indicated that nitrate was mainly reduced in its logarithmic growth phase with a very low leve1 accumulation of nitrite,suggesting that the aerobic denitrification process of strain S1-1 occurred mainly in this phase.The GC-MS results showed that N 2 O was formed as the major intermediate during the aerobic denitrifying process of strain S1-1.Finally,factors affecting the growth of strain S1-1 and its aerobic denitrifying ability were also investigated.Results showed that the optimum aerobic denitrification conditions for strain S1-1 were sodium succinate as carbon source,C/N ratio15,salinity 10 g/L NaCl,incubation temperature 20°C and initial pH 6.5. 展开更多
