METHODS: Tolerance towards human...AIM: To determine whether normal genetically immunocompetent rodent hosts could be manipulated to accept human hepatocyte transplants with long term survival without immunosuppression. METHODS: Tolerance towards human hepatocytes was established by injection of primary human hepatocytes or Huh7 human hepatoma cells into the peritoneal cavities of fetal rats. Corresponding cells were subsequently transplanted into newborn rats via intrasplenic injection within 24h after birth. RESULTS: Mixed lymphocyte assays showed that spleen cells from non-tolerized rats were stimulated to proliferate when exposed to human hepatocytes, while cells from tolerized rats were not. Injections made between 15 d and 17 d of gestation produced optimal tolerization. Transplanted human hepatocytes in rat livers were visualized by immunohistochemical staining of human albumin. By dot blotting of genomic DNA in livers of tolerized rats 16 weeks after hepatocyte transplantation, it was found that approximately 2.5 X 10(5) human hepatocytes survived per rat liver. Human albumin mRNA was detected in rat livers by RT-PCR for 15 wk, and human albumin protein was also detectable in rat serum. CONCLUSION: Tolerization of an immuno-competent rat can permit transplantation, and survival of functional human hepatocytes.展开更多

In this essay,our goal is to discover science in Martin Heidegger’s Introduction to Metaphysics,lecture notes for his 1935 summer semester course,because,after all,his subject is metaphysica generalis,or ontology,and...In this essay,our goal is to discover science in Martin Heidegger’s Introduction to Metaphysics,lecture notes for his 1935 summer semester course,because,after all,his subject is metaphysica generalis,or ontology,and this could be construed as a theory of the human brain.Here,by means of verbatim quotes from his text,we attempt to show that indeed these lectures can be viewed as suggestion for an objective scientific theory of human perception,the human capacity for deciphering phenomena,i.e.hermeneutics in its broadest sense.His added notes from the 1953 edition,all of which are comments,not corrections,imply that he never abandoned these thoughts on metaphysics,despite all of his utterances about a need to overcome it,and their popular interpretations to that effect.In his presentation,he further develops the colorful and intuitive style,an hermeneutic language,that he had created in his earlier work Being and Time.The logical functions of Dasein’s anatomical brain are performed by the logos machine,formerly the human soul,using top-down processing based on a global context,the noumenal cosmos which humans maintain internally.Heidegger’s 1942/43 winter semester lectures Parmenides extend in unbroken fashion his 1935 work,proving that he never abandoned,as is widely claimed,his metaphysical avenue of thought.展开更多

Each fit function is expressed as a temperature function in terms of a power series expansion about the critical point. The leading term with the critical exponent dominates the temperature variation between the critical and triple points. With β being introduced as the critical exponent for the difference between liquid and vapor densities, it is shown that the critical exponent of each fit function depends (if at all) on β. In particular, the critical exponent of the reciprocal heat capacity c﹣1 is α=1-2β and those of the entropy s and internal energy u are?2β, while that of the reciprocal isothermal compressibility?κ﹣1T is γ=1. It is thus found that in the case of the two-phase fluid the Rushbrooke equation conjectured α +?2β + γ=2 combines the scaling laws resulting from the two relations c=du/dT and?κT=dlnρ/dp. In the context with c, the second temperature derivatives of the chemical potential μ and vapor pressure p are investigated. As the critical point is approached, ﹣d2μ/dT2 diverges as c, while?d2p/dT2 converges to a finite limit. This is explicitly pointed out for the two-phase fluid, water (with β=0.3155). The positive and almost vanishing internal energy of the one-phase fluid at temperatures above and close to the critical point causes conditions for large long-wavelength density fluctuations, which are observed as critical opalescence. For negative values of the internal energy, i.e. the two-phase fluid below the critical point, there are only microscopic density fluctuations.
